Objective To detect the expression of Wnt5a, β-catenin, E-cadherin genes and proteinsin gastric cancer cell line SGC7901and normal gastric cell line GES-1, combined with theexpression of Wnt5a in gastric carcinoma and adjacent normal tissue distal to explore thefunction and the relationship between occurrence and development of gastric cancer, as wellas the roles of Wnt5a, β-catenin and E-cadherin in cell signal transduction, to bring new hopefor the treatment of gastric cancer in clinical immunology, to establish new and more effectivetreatments. Methods Culture normal gastric mucosa cell lines GES-1and gastric cancercell lines SGC7901in vitro. The expression of Wnt5a, E-cadherin, β-catenin mRNA in GCcell lines and normal gastric mucosa cell lines was detected by Real-time reversetranscription-polymerase chain reaction (RT-PCR). The expression of Wnt5a, β-catenin,E-cadherin proteins in GC cell lines and normal gastric mucosa cell lines was detected byWestern-blot. Collected55cases of Gastric cancer samples from clinical withoutchemotherapy and radiotherapy, then took tissue classification and clinical stage. At the sametime took28cases tumor-adjacent normal tissue samples (both confirmed no cancer cells)as normal control, conventional paraffin embedding and sectioning. The expression of Wnt5aproteins in55GC and28matched tumor-adjacent normal tissue samples were examined byimmunohistochemistry using the streptavidin-peroxidase method. Results (1)The expressionof Wnt5a, β-catenin, E-cadherin mRNA and proteins were be detected in normal gastricmucosa cell lines GES-1and gastric cancer cell lines SGC7901by RT-PCR andWerstern-blot.(2)The relative level of Wnt-5a, β-catenin mRNA and Wnt-5a, β-cateninproteins were both significantly higher in GC cell lines SGC7901than in normal gastric mucosa cell lines of GES-1detected by Realtime-PCR and Western-blot (P<0.05), while thelevel of E-cadherin mRNA and E-cadherin protein are significantly lower in SGC7901celllines than in GES-1cell lines (P<0.05).(3)The rate of Wnt-5a protein expression was alsosignificantly higher than the normal tissue samples (6/28, P<0.05). The expression of Wnt5awas associated with higher degree of regional lymph node metastasis and later TNM stage (P<0.05). Conclusion (1)The expressions of Wnt5a, β-catenin, E cadherin are consistent in thelevel of transcription and translation.(2) Wnt5a, β-catenin genes play an oncogene-like role inthe development of gastric carcinoma, and E-cadherin gene play the role of tumor suppressorgenes in the development of gastric carcinoma.(3)Positive expression of Wnt5a wasassociated with higher degree of regional lymph node metastasis, later TNM stage, and higherrate of female patients (P<0.05).
