| Objective：Rheumatoid arthritis (RA) which is a chronic, symmetrical polyarthritisis anautoimmune disease characterized by joint destruction. Immune dysfunction, synovialcell accumulation and synovial thickening, vascular hyperplasia of articular cartilageand bone destruction is the main pathological link, but its pathogenesis is still unclear.Clinical has no cure for the disease, and now the clinical medication main purpose isto improve the clinical symptoms, delay the progression of the disease, reduce jointdamage and improve the life quality of the patients. Drugs currently in clinicaltreatment of RA mainly include two categories: improving clinical symptom classantirheumatic drugs and improve the condition class antirheumatic drugs.Antirheumatic drugs of Improving the symptoms for clinical is easy to relapse afterdrug withdrawal and have long-term side effects. Disease modifying drugs can delaythe progression of the disease, but in the use process there are many adverse effects,and can prevent the deterioration of disease. In order to seek the effective cure for thedisease, a large number of medical scientists have done a lot of theoretical andexperimental researches on traditional Chinese medicine treatment of the disease. It isreliable that the motherland medicine curative effect in the treatment of RA haveobvious advantages, especially in the reduction of Western medicine side effect,reducing the recurrence rate, improving the curative effect and so on, which showsgood prospects. Toddalia asiatica is widely used as one of the drug in the treatment ofrheumatoid disease inTujia area, adjuvant arthritis in this experiment is based onFreund’s complete adjuvant induced (adjuvant arthritis, AA) rat model, thepreliminary mechanism through observing the Toddalia asiatica expression of ethanoland water extracts on improvement of AA rats ankle swelling and observation onsynovial biopsy to evaluate the treatment of rheumatoid arthritis, and the role of expression and synovial IL-1detection of rat serum β, TNF-α, IL-8VEGF, MMP3isto discuss the treatment of adjuvant arthritis rats.Methods：1. To establish the model of adjuvant arthritis rats.2. To observe arthritis swelling and the paw volume determination.3. TodetecteInfluence of synovial tissue of rats by Toddalia asiatica.4. Todetectethe level of serum IL-1β, TNF-α, IL-8of the secretion of rats ineach group with ELISA method.5. Todetecte of the expression levels of synovial tissue immunohistochemistrystaining of VEGF, MMP-3.Result：1. It is successful to establish the model of adjuvant arthritis in rats. The rats inthe model group appeared the joint swelling, tail nodules, ears red spots and nodules,foot swelling rate was significantly increased (p<0.05).2. Toddalia asiatica great improves ankle swelling of AA rats, has significantdifference with the model group (p<0.05); from the pathological sections, it isobvious that compared with the model group the inflammatory and cell secretionhas reducedandcell proliferation, angiogenesis is not obvious.3. Toddalia asiatica decreased the expression of serum IL-1, TNF-α, IL-8andthere are significant differences compared with the model group (p<0.05).4. The secretion of rats in each group were detected the level of serum IL-1β,TNF-α, IL-8ELISA method.5. The detection of the expression levels of synovial tissueimmunohistochemistry staining of VEGF, MMP-3.Conclusion：1. Toddalia asiatica on the rat model of AA has a therapeutic effect, improves thejoint swelling in rats, decreases the secretion of inflammatory cells in the synovium, inhibiting angiogenesis, and reduces cell proliferation.2. Expression of Toddalia asiatica through the inhibition of IL-1, TNF-α, IL-8proinflammatory factor reduces the secretion of inflammatory cells and improvesarthritis symptoms relieve joint swelling, which may be a mechanism for thetreatment of RA.3. Expression of Toddaliaasiatica possible through inhibition of VEGF,MMP-3reduces vascular hyperplasia, cartilage degradation and improve the conditionof RA.The results suggested that the Toddalia asiaticahas the treatment of rheumatoidarthritis, inhibite inflammatory factors secretion. Reduction angiogenesis in synovialmembrane and cartilage destruction is its possible mechanism, which Provides thebasis for the advanced study of mechanism of action for the Toddalia asiatica in thetreatment of RA. |
