Cerebral infarction is the most frequent diseasee in nerve system. It threats humanhealth severely. When damage of cerebral vessels in stroke happens, the EPCs can cometogether to form new blood vessels at damaged or ischmic tissue selectively.The studies ofischemic diseases of heart,brain,liver and kidney increase day by day,have become hotspotof study.Objective: Inject EPCs into rsts with acute cerebral ischemia-reperfusion model andinvestigate the effet of EPCs on the treatment of acute cerebral ischemia disease.Materials and Methods:1. Mononuclear cells were obtained by using density gradientcentrifugation and adherence screening method from bone marrow and induct them,observedand recorded the biological characteristics of cells. EPCs specific surface markers KDR,CD34is selected and detected by immunofluorescence.2.The acute cerebral ischemia-reperfusion in rats of left middle cerebral arteryocclusion(MCAO)was made by suture method. Sixty male Wistar rats weighing250to300g were divided into three groups randomly: sham group12(group A), PBS-treated group20(group B), EPCs-treated group28(group C). Brains were taken and detected at1d,3d,7dand14d according to different time after transplanting EPCs.(1)The neurological behavior scores were evaluated on MCAO rats after operation, andthe rat which was1-3score was selected into follow experiment.(2)Observe the scope of cerebral infarction by TTC staining.(3)Histopathologic changes of brains was observed by HE staining.(4)The expression of VEGF、FGF-2of different time in rats was determined by meansof immunohistochemistry. The positive cells were observed and recorded by microscope.The results were analyzed through SPSS17.0software.Results:(1)EPCs were isolated from bone marrow and cultured successfully.(2)Attached cells were positive for CD34, KDR by immunofluorescence,(3)Acute cerebral ischemia-reperfusion in rats of middle cerebral artery occlusion wasmade by suture occlusion method. (4)The neurological scores in group C were lower than group B, especially after7days oftreatment.(5)TTC staing: Total infarct volume in group C were reduced significantly compared togroup B(P<0.05).(6)Expression of VEGF、FGF-2: The expression of VEGF and FGF-2in group Cincreased obviously than other two groups.Conclusion:(1)EPCs can be isolated from bone marrow of rats,cultured and identifiedsuccessfully,will offer foundation to the therapy of ischemic cerebrovascular disease.(2)Treatment of acute ischemic stroke with EPCs has good effect, can promoteangiogenesis,save ischemic penumbra,decrease infarction volume, improve neurologicaldeficits and the recovery of injured nerve.
