Objective:The chronic cerebral ischemia rats at different time points and MCP-1expression in brain tissue changes and relationship with chronic cerebral ischemiainduced vascular dementia.Methods:Choose48Wistar male rats were randomly divided into model group, ischemia(15days after operation,1months,2months group), sham-operation controlgroup.Through the bilateral common carotid artery ligation in permanently (2VO)animal model of chronic cerebral ischemia; Application of Morris water maze test onlearning and memory in rats at different time points of various groups feature;Lightmicroscopic observation of pathological morphology of hippocampal CA1area ofrats in each group changes by HE staining;Using enzyme-linked immunosorbentassay (enzyme-linked immunosorbent assay ELISA) to detect the expression of ratMCP-1of each category.Observe the histological changes in behavior andpathology after chronic cerebral ischemia in rats.To explore the possiblerelationship between MCP-1and chronic cerebral ischemia induced vasculardementia.Results:The groups of chronic cerebral ischemia in rats by the water maze test its spatiallearning and memory capabilities are obstacles,and with the extension of ischemictimecognitive impairmentis more obvious.There were significant differencescompared with control group(P<0.05).After HE staining to observe the pathologicalchanges:Sham operation group, the number of species distribution of hippocampalCA1pyramidal cells, cells closely arranged neatly, the nucleus and the cell structureintegrity, cytoplasm is homogeneous, clear nucleolus.The CA1area of hippocampusneuralcellsin ischemic group arranged disorderly,irregular shape,nuclearpyknosis,karyorrhexis, cytoplasmic inhomogeneity,nucleolaruncleardisplay.Different degrees of degeneration,necrosis of neurons visible can see, the longer of Ischemia time,the more obvious of this changes.The results of ELISAshowed that the expression of MCP-1are increased significantly in rats of eachgroup after2VO compared with the sham operation group,there are statisticallysignificant(P﹤0.05). The increased expression of MCP-1participate in the processof vascular dementia induced by chronic cerebral ischemia.Conclusion:1.Permanent bilateral common carotid arteries ligation method (2vo) producedby chronic cerebral ischemia model can lead to progressive cognitive dysfunction inrats,the model is reliable.2.The rats hippocampal CA1area appear advancedpathological damage After chronic cerebral ischemia,and this kind of damage andprogressive cognitive dysfunction is parallel.3.Chronic cerebral ischemia can cause highcontinuous expression of MCP-1in rat brain.
