3-n-butylphthalide (NBP) was synthesized and developed as an anti-cerebral ischemia agent. It was approved by SFDA of China in September 2002. The previous studies showed that NBP was a potentially beneficial and promising drug for the treatment of stroke with multiple actions which affect different pathophysiological processes. NBP is a chiral compound, and contains 1- and d- isomers. Recently, 1-NBP and d-NBP were separated, but previous research found that there was antagonistic action between 1- and d-NBP. The present research was designed to investigate the effects and mechanisms of 1-NBP on transient cerebral ischemia, platelet aggregation and experimental senile dementia.PART I Effects of 1-NBP on cerebral infarct volume after middle cerebral artery occlusionIn the present study, we administered 1-, d- and dl-NBP orally to rats and investigated the effects on ischemic brain damage by middle cerebral artery occlusion (MCAO). The infarct volume and behavioral scores were dose-dependently decreased by 1-NBP. The protective effect was similar with MK-801. But, d- and dl- NBP had less effect on cerebral infarct volume and behavioral test. Furthermore, 1-NBP 20 and 40 mg/kg were more potent than dl-NBP 40 and 80 mg/kg. The research indicated that d-NBP maybe antagonize 1-NBP on neuroprotection.
