Objective To investigate the effects of1,25-(OH)2VD3on GATA-3/T-bet and IL-33in asthma model, to explore the mechanism of1,25-(OH)2VD3in the treatment of asthma.Methods Thirty-two female Kunming mice were randomly divided into4groups:the control group (group A), the asthma group (group B), the dexamethasone treated group (group C) and the1,25-(OH)2VD3treated group (group D). The allergic mouse models were established by ovalbumin (OVA). Group C and group D were given different therapy. Observed the general situation of each mouse. The number of leukocytes and the eosnophils in the bronchoalveolar lavage fluid (BALF) were counted. The histological changes of lung tissue were measured after HE staining. Enzyme-linked immunosorbent assay (ELISA) was used to detecte the levels of IgE and IL-33in the serum. The expressions of transcription factor GATA-3and T-bet protein in the lung were measured by immunohistochemical analysis.Results1The number of total leukocytesv and asinophil population in BALF of groups B were higher than other groups (all P<0.01), and that in group A were the lowest in the four groups (all P<0.01).There were significant differences between any two groups besides group C and group D.2The levels of IgE in serum of group B were the highest in the four groups while that in group A were the lowest, and there were significant differences (all P<0.01). There were significant differences between any two groups (all P<0.01), but there was no significant differences between group C and group D(P>0.05).3The level of IL-33were significantly higher in group B than that in the other three groups (all P<0.01). That was distinctly lessoned in group C and D compared with group B(all P<0.01), but no significant result was observed between group C and group D (P>0.05).4Immunohistochemecal results revealed:The expression of GATA-3protein in group B were siginificantly enhanced than other there groups (all P<0.01).No significant differences was detected between group C and group D (P>0.05). The expression of T-bet protein in group B were siginificantly lowered contrasted with group A and group D (all P<0.01). However, there were no significant improvement in group C when compared with group B (all P>0.05).5The correlation analysis revealed that IL-33level was a siginificantly positive correlation with IgE(P<0.01). Conclusions1Our study shows that the imbalance of the T cell-specific transcription factor GATA-3and T-bet contributes to the process of asthma.2Dexamethasone can suppress the inflammation of asthma by down-regulating the expression of GATA-3and increasing the expression of T-bet, while1,25-(OH)2VD3involves in the inflammation process of asthma by decreasing the GATA-3level.3IL-33can promote the airway inflammation of asthma,which can be restained by1,25-(OH)2VD3.4.1,25-(OH)2VD3can relieve the respiratory inflammation in acute asthmatic mice, which highlight the road of asthma therapy.
