ObjectiveDiscussion isolated and cultured canine bone marrow mesenchymal stem cells andosteogenic differentiation of methods and results..To evaluate the effect of bone marrowmesenchymal stem cells combined with calcium phosphate cement scaffold in bonetissue regeneration.Provide new ideas and experimental basis for the repair andreconstruction of oral implants bone tissue defects.MethodsIsolated and cultured canine bone marrow mesenchymal stem cells,observed theosteogenic induction effect,then combined cells with calcium phosphate cement scaffoldfor animal experiment.In four beagles create three certain size bone defects on each sideof the mandible.The bone defects were randomly divided into threegroups:BMSCs-CPCgroup(scaffolds transplanted composite seed cells),CPC group(onlygraft scaffold),control group(no treatment).After transplantation4weeks and8weeks,two dogs were sacrificed,respectively.Clinical observation, X-ray imaging,histologic analyses were performed to evaluate the level of bone defect repairing indifferent periods.ResultsAfter14days,bone marrow mesenchymal stem cells into osteoblasts inducedcalcification in nodule formation; After4,8weeks,bone tissue regeneration of BMSCs-CPC group and CPC group was better than the control group.The maturity ofnew bone tissue,scaffold degerdation, bone maturation in the BMSCs-CPC group wasbetter than the CPC group.ConclusionMesenchymal stem cells are a multipotent differentiation potential and highproliferative capacity of stem cells that can differentiate into osteoblasts.Calciumphosphate cement is a kind of bioactive scaffold for maintaining bone shape toaccelerate osteogenesis play an important role.BMSCs seed cells combine with CPCscaffolds materials repair bone defects is an effective,practical method.For the late oralimplantology provided a useful repair condition.
