| Purpose: To explore the differential proteome pattern in fibrosis liver in comparison towild type. Method: Mice were fed for15weeks with olive oil or carbon tetrachloride.Mouse livers from both groups were collected and submitted to MS platform forproteome screening. GO (Gene Ontology) biological process and KEGG (KyotoEnyoolpedia of Genes and Genomes) pathway enrichment analysis were employed toanalyze differentially expressed proteins. Results: In both groups, we identified17382and20486unique peptides in control and carbon tetrachloride-induced groups, respectively. A total of4,991proteins were identified, and2135of which weredifferentially expressed.1264proteins were up regulated and871proteins were downregulated. Conclusion: Proteins associated with DNA replication, cell cycle,ECM-receptor interaction, and splicesome are significantly enriched in carbontetrachloride-induced group. Those findings showed that fibrosis is closely related toregulation of VEGF and T cell receptor signaling pathway, suggesting liver fibrosis isa complex signal transduction process that many biological process such as livermetabolism, inflammation, and immune response are involved, the function of proteinmetabolism in liver parenchymal cells and blocking the transduction of inflammatorysignaling path-way may be beneficial for liver fibrosis therapy. |
