Begonia grandis Dry. as an traditional Chinese medicine originated from Genus Begonia plants. The crude drug was harvested in late summer or early autumn. Borning in wet forest or gutter and distributing the provinces south of the Changjiang, north to Shandong, Hebei with abundant wildlife resources. It was recorded that this medicine was used to treat bruises, vomiting, diarrhea, irregular menstruation, uterine bleeding, vaginal discharge, pouring muddy, sore throat, etc. It have been used as a folk medicine for a long time. However, the research on anti-inflammatory ingredients and Pharmacognosy of the plant is almost empty. In this paper, the Pharmacognosy of Begonia grandis Dry. were investigated comprehensively. Meanwhile, preliminary pharmacodynamics of anti-inflammatory active fractions of Begonia grandis Dry. were explored. There is a good broad application and promising development prospect.The study of macroscopic and microscopic identification showed that vascular is a ring due to the pericycle fiber and the bundle fiber are connected in the tubers of Begonia cortical, column and pith have not clear boundaries in petiole.16to19vasculars form a ring intermittently. The leaves show multicellular non-glandular hairs and multicellular glandular hairs, infinitive of blowhole,3to4subsidiary cells. the whole plant powder including that prismatic crystals of calcium oxalate, non-glandular hairs, vessel are the main characteristics. These results had significant value to provide a reference for the identification of Begonia grandis Dry.Using liquid chromatography electrospray ionization mass spectrometry, established the fingerprint of flavonoids and organic acid. Furthermore,9compounds were identified from the fingerprint peaks. There are6compounds isolated from this plant for the first time.The determination method of total flavonoids was established by UV spectrophotometry. The results show that its contents of total flavonoids were different significantly in different origin and different parts. The content of total flavonoids in tubers is higher than in stems, then higher than in leaves. The samples collected in October contained higher content of total flavonoids than that in August, then higher than in June. The samples collected in Yichang contained higher content of total flavonoids than that in Enshi. Therefore, it was preliminarily considered that this crude drug could be collected in Autumn.The determination method of flavonoids and organic acid ingredients was established by UPLC, The results show that its contents were different significantly in different origin and different parts. The content of3,4-dihydroxybenzoic acid, protocatechuic aldehyde, catechin and epicatechin in tubers are higher than in leaves, then higher than in stems, which are little to do with the harvest season and the different of origin. Differently, the content of rutin in leaves is higher than in stems, then higher than in tubers. The samples collected in Yichang contained higher content of rutin than that in Enshi. Such results supply a better basis for its harvesting and the quality control.RAW264.7macrophage line in mice was induced by LPS to set up the inflammatory model. ELISA assay was used to measure the secretion of TNF-a, IL-1β and NO in the supernatant of drug treated and untreated macrophages. The results showed that the ethanol extracts, ethyl acetate extracts and butanol extracts from Begonia grandis Dry. had similar effects on inflammation as Dexamethasone, it could inhibit inflammation by down regulating the expression and secretion of pro-inflammatory factors.
