Objective: To investigate the significance of MMP-7mRNA, hTERT expression in theplasma and combined detection of serum CEA, CA19-9, CYFRA211, SCC, NSE andProGRP levels in lung cancer patients;to compare sensitivity of the three results. In a futurefollow-up, to discuss the relationship between MMP-7mRNA, hTERT, serum CEA andrelated proteins with lung cancer prognosis.Method: Analyze158cases of clinical information and follow-up survival with untreatedlung cancer patients between September2010-2013September in the department ofrespiratory in Shan Xi Province Tumor Hospital. Peripheral blood samples were obtainedfrom158lung cancer patients before treatment and104benign lung disease patients(controlgroup) diagnosed at the same time.MMP-7mRNA and hTERT in the plasma were measuredby RT-PCR;serum CEA, CA19-9, CYFRA211, SCC, NSE and ProGRP levels weremeasured by electrochemiluminescence immunoassay(ECLIA);Serum CEA, CA19-9,CYFRA211, SCC, NSE and ProGRP levels, MMP-7mRNA and hTERT expression in lungcancer were compared using the X2test;Survival analysis using Kaplan-Meier method,significance test of Log-rank method.Results:①The positive rate (77.2%)of combined detection of CEA and related proteins inlung cancer patients were significantly higher than those of MMP-7mRNA(48.1%),hTERT(53.2%) in the plasma and benign lung disease patients(control group)(23.1%)(P<0.05).②The diagnostic positive rate of combined detection of CEA, CA19-9, CYFRA211, SCC,NSE and ProGRP levels were significantly higher than those of MMP-7mRNA and hTERTfor clinical staging, lymphatic metastasis and pathological type(P<0.05).③Follow-up results show, Median survival times(MST)of total negative、single positive、two positive、three positive、four positive and five positive from the results of serum CEAand related proteins were respectively30months、22months、12months、18months、16months、28months(P<0.003).④At the follow-up visit, combination of total negative and single positive from the resultsof serum CEA and related proteins was named group1, combination of more than twopositive from the results of serum CEA and related proteins was named group2.Median survival times(MST) of the two groups were respectively29months and15months.(P<0.001) ⑤At the follow-up visit, Median survival times (MST) of total negative ofMMP-7mRNA、hTERT expression, total positive of MMP-7mRNA、hTERT expressionand any positive of MMP-7mRNA、hTERT expression were respectively21months,21months,26months,23months。(P>0.05)Conclusion: The sensitivity of combined test for CEA, CA19-9, CYFRA211, SCC, NSEand ProGRP levels were superior to those of MMP-7mRNA and hTERT. And the positiverate of CEA, CA19-9, CYFRA211, SCC, NSE and ProGRP levels were higher than those ofMMP-7mRNA and hTERT related to clinical staging, lymphatic metastasis and pathologicaltype. The number of positive and detecting concentration from serum CEA and relatedproteins were closely related to lung cancer prognosis. There was no significant correlationbetween combined detection of MMP-7mRNA、hTERT and lung cancer prognosis. Itcan be seen as simple but reliable method for diagnosis, monitoring recurrence of lungcancer.
