1ObjectiveTo observe the influence of Yiqi Jiedu Huayu Recipe on ulcerative colitis (UC)induced by trinitrobenzene sulfonic acid (TNBS)/ethanol method of the colonicepithelial barrier in rats experiment, detection of rat serum interleukin-23(IL-23),interleukin-17(IL-17), interleukin-10(IL-10) and colonic mucosa filamentous actinexpression, and to explore the effects of experimental rats in colonic epithelial barrierand its therapeutic mechanism of Yiqi Jiedu Huayu Recipe on ulcerative colitis (UC).2MethodIn52SD rats randomly take10only as normal group, the remaining42only bytrinitrobenzene sulfonic acid (TNBS)/ethanol ulcerative colitis model rats. Randomlyselected two model rats to be put to death, only take colon pathological sliceobservation, confirmed UC experimental model in rats. The remaining40wererandomly divided into model group, Yiqi Jiedu Huayu Recipe high and low dosegroup and Salicylazosulfapyridine (SASP) group, continuous lavage treatment for2weeks. To observe inflammatory activity index (DAI), gross morphology damageindex (CMDI) and changes in histological damage index (TDI) in each rat betweengroups, to observe pathological morphology of colon tissue and the changes of theultrastructure of colonic mucosa, and with ELISA method in rats serum IL-23, IL-17,IL-10levels and using immunohistochemical method to observe the influence of thecolonic mucosa filamentous actin expression.3Result3.1Each treatment group could improve the symptoms of UC rats, With Yiqi JieduHuayu Recipe high dose group and SASP group is more obvious.3.2Compared with model group, Yiqi Jiedu Huayu Recipe each dose group and SASPgroup rats DAI, CMDI, TDI scores were significantly lower, all have significant difference (P<0.01). Compared with low dose group, high dose group and SASPgroup of DAI, CMDI, TDI were with statistical significance (P<0.05).3.3Compared with model group, IL-10in each treatment group was obviously higher,IL-23, IL-17levels decreased obviously(P<0.01or P<0.05) Compared with SASPgroup, Yiqi Jiedu Huayu Recipe high and low dose group of IL-10levels increasedsignificantly(P<0.01or P<0.05); Compared with Yiqi Jiedu Huayu Recipe low dosegroup, high dose group significantly increased levels of IL-10, IL-23, IL-17decreased significantly(P<0.01).3.4Compared with model group, colonic epithelium in each treatment group ratsconnected between density, closely connected structure integrity, width of cell gapsignificantly reduced, bridge structural integrity, and no expansion of endoplasmicreticulum and mitochondria swelling.3.5Compared with model group, the expression of F-actin protein in each treatmentgroup rats colon tissue were significantly increased(P<0.01); Compared with SASPgroup, low dose group have significant differences(P<0.05), and high dose group, nosignificant differences(P>0.05); Compared with Yiqi Jiedu Huayu Recipe low dosegroup, the expression of F-actin protein of high dose group rats intestinal tissueincreased significantly(P<0.01).4Conclusion4.1Yiqi Jiedu Huayu Recipe could obviously improve the symptoms of UC modelrats.4.2Yiqi Jiedu Huayu Recipe can obviously reduce in the UC model rats of DAI,CMDI, TDI.4.3Yiqi Jiedu Huayu Recipe can increase the UC model rats serum IL-10levels,reduce the serum IL-23, IL-17levels.4.4Yiqi Jiedu Huayu Recipe can improve colonic mucosa of the UC model rats byelectron microscopy ultrastructure.4.5Yiqi Jiedu Huayu Recipe increase colonic tissue in the UC model rats of the expression of fiber filamentous actin.
