Objective:By observing the effect of Xingqitiaoshen acupuncture therapy on expressioncAMP response element binding protein(CREB) and phosphodiester4(PDE4)in leftfrontal cortex and hippocampus in post-stroke depression(PSD) rats so as to explore itsmechanisms underlying improvement of PSD in clinical acupoint selection.Methods:1.Selecting eighty male sprague dawley (SD) rats were health,which is7-8months yearsold and weight300g±50g.Doing the open-Field test after adaptive feeding one week,therating similar rats were randomly divided into normal control(n=17),and the other ratswere prepared cerebral ischemia model by coagulating middle cerebral artery,thenfifty-one rats had been passed Zea Longa neurobehavioral test.They were randomly dividedinto model,acupuncture and western medicine groups(n=17in each group).During theexperiment has complied with the rules of3R strictly.seven kinds of chronic stressmethods combined with separation to produce the depressive model while doing ZeaLonga neurobehavioral test. The8th day of chronic stress start to treat,and acupuncturewas applied to bilateral “Renzhong”(DU26),“Hegu”(LI14)and “Taichong”(LR3)for15min,once daily for42sessions (standard according to the seventh edition ExperimentalAcupuncture and Moxibustion)for acupuncture group.The western medicine group withfluoxetine hydrochloride capsule be gavage by1mL/100g for once a day while theacupuncture group treat.Besides,model and normal control groups used0.9%NS to begavage by1mL/100g for once a day,which has the same treatment with acupuncturegroup.2.Zea Longa neurobehavioral test and Open-Field test of horizontal motion and verticalmovement were detected in the first day of chronic stress,treatment the21th days and42th days.In the second day,9th day,16th day and23th day,groups rats were observedthe consumption of sucrose water in chronic stress.In the end of chronic stress,the PSDmodel rats begin to detect the Morris water maze test to observe the ability of learning and space exploration.Taken immediately after the end of treatment,using RT-PCRdetected the expression of CREBmRNA and PDE4mRNA in the left frontal cortex andhippocampus.Results:1.Compared with western medicine group,there was no significant difference betweenthe acupuncture group and western medicine group for the consumption of sucrosesolution, horizontal motion scores,space exploration ability of the Morris water mazetest,the expression of CREBmRNA in the left frontal cortex and the expression ofPDE4mRNA in hippocampus(P>0.05).But there was significant difference between theacupuncture group and western medicine group for vertical movement scores,learningability of the Morris water maze test,Zea Longa neurobehavioral test,the exPression ofCREBmRNA in hippocampus and the expression of PDE4mRNA in the left frontalcortex(P<0.01orP<0.05).2.Compared with normal control group,efficacy of western medicine group was not asgood as it in the consumption of sucrose solution,horizontal motion scores and verticalmovement scores, the ability of learning and space exploration of the Morris water mazetest and Zea Longa neurobehavioral test(P<0.01orP<0.05).The expression level ofCREBmRNA in western medicine group was up-regulated significantly comparing withnormal control(P<0.01orP<0.05),while the expression level of PDE4mRNA in leftfrontal cortex and hippocampus of western medicine group was significantly lowcomparing with normalcontrol(P<0.01).The consumption of sucrose solution,horizontalmotion scores and vertical movement scores,the ability of learning and spaceexploration of the Morris water maze test and Zea Longa neurobehavioral test of themodel group was inferior to normal group(P<0.01).The expression levels ofCREBmRNA in the left frontal cortex and hippocampus were down-regulatedsignificantly comparing with normal control(P<0.01),and the expression levels ofPDE4mRNA in left frontal cortex and hippocampus of model group were significantlyhigher than normal group(P<0.01).Compared with normal control group,acupuncture group was not good as it in the consumption of sucrose solution,horizontal motionscores and vertical movement scores,space exploration ability of the Morris water mazetest and Zea Longa neurobehavioral test(P<0.01orP<0.05),but there was no significantdifference between normal group in learning ability of the Morris water mazetest(P>0.05). In comparison with normal group,the expression level of CREBmRNA inacupuncture group was up-regulated significantly in the left frontal cortex andhippocampus(P<0.05),and the expression levels of PDE4mRNA in hippocampus ofacupuncture group down-regulated significantly(P<0.01),but there was no significantdifference between normal group for the expression level of PDE4mRNA in the leftfrontal cortex(P>0.05).3.Compared with model group,acupuncture group and western medicine group weresuperior to it in the consumption of sucrose solution,horizontal motion scores andvertical movement scores, the ability of learning and space exploration of the Morriswater maze test and Zea Longa neurobehavioral test,there were significant differencebetween model group(P<0.01orP<0.05).The expression level of CREBmRNA in theacupuncture group and western medicine group were up-regulated significantly,whichhas significant difference between model group(P<0.01).Conversely,the expressionlevel of PDE4mRNA in the acupuncture group and western medicine group weredown-regulated significantly,which has significant difference between modelgroup(P<0.01).Conclusion:Xingqitiaoshen acupuncture therapy has a mechanism that is excitation theexpression of CREBmRNA and inhibition the expression of PDE4mRNA to treat PSD.
