Objective:To observe the expression of CREBmRNA and PDE4mRNA and the interference effect of Yinao Jieyu prescription on the ratmodel of ischemic post-stroke depression.Method:Male Wister rats were randomly divided into normal group, false surgical group, model group, Chinese traditional medicine treatment group and Western medicine control group according to the scores of OPEN-FIELD test. Rats in the normal group received no intervention. Those in other four groups were duplicated into focal ischemic first, but the rats in the false surgical group only open up the left skull and uncover the duramater, and they were made into PSD model.And then they received gastric perfusion of double distilled water, fluoxetin and Yinnao Jievu prescription respectively once a day for 6 weeks from the 1st day after the establishment of model. After that, Reverse transcription polymerase chain reaction (RT-PCR) was used in the ratmodel of the five groups. The expression of CREBmRNA and PDE4mRNA in prefrontal cortex and hippocampus, and the interference of Yinao Jieyu prescription were observed.Result:The expression of CREBmRNA in hippocampus and prefrontal cortex in the ratmodel of ischemic post-stroke depression in the model group decreased obviously compared to that of the normal group, while Yinao Jieyu prescription improved obviously the expression of CREBmRNA. The expression of CREBmRNA in the brain of Rats in the false surgical group depression decreased. The expression of PDE4mRNA in hippocampus and prefrontal cortex in the ratmodel of ischemic post-stroke depression in the model group increased obviously compared to that of the normal group, and the expression in hippocampus is higher than that in prefrontal cortex clearly. Yinao Jieyu prescription improved obviously the expression of PDE4mRNA, especially in hippocampus. The expression of PDE4mRNA in the brain of Rats in the false surgical group increased.Conclusion:The result showed that the effect of Yinao Jieyu prescription is related to its function of adjusting the expression of CREBmRNA and PDE4mRNA.
