Objective Establish the model rats acutely stressed to plateau. To investigate the change of autophagy after the intestinal injury by hematoxylin-eosin staining, transmission electron microscopy and immunohistochemistry, and the result may provide theoretical basis for the role of autophagy with intestinal disorders in plateau area in further research.Methods Fifty Wistar rats were randomly divided into five groups:the plain control group, the acutely plateau6h,12h,24h and48h group, each consisting of10rats. The acutely plateau groups were established the model rats exposed to hypobaric hypoxia at a simulated altitude of4767m by decompression chamber. The injury of the intestinal was observed by light microscopy. The autophagosome in the intestinal epithelial cells were observed by transmission electron microscopy. The expression of Beclinl and LC3B protein were analyzed by immunohistochemistry.Results Light microscopy shows:the intestinal mucosa may mild damage in the acutely plateau6h group compared with the plain control group, and the damage obvious aggravate in the acutely plateau12h and24h group. It was the most serious in the acutely plateau48h group. Transmission electron microscopy shows:the double membrane structure of autophagosome began to find in the acutely plateau6h group, and increased the most obviously in the acutely plateau24h group. It is still observed in the acutely plateau48h group. Immunohistochemistry shows:the expression of Beclinl in the plain control group, the acutely plateau6h,12h,24h and48h group were (0.142±0.005),(0.161±0.017),(0.179±0.018),(0.195±0.015),(0.185±0.020), and the expression of Beclinl in the acutely plateau6h,12h,24h and48h group were significantly higher than that in the plain control group (P<0.01). The expression of LC3B in the plain control group, the acutely plateau6h,12h,24h and48h group were (0.150±0.007),(0.171±0.015),(0.190±0.017),(0.207±0.016),(0.193±0.019), and the expression of LC3B in the acutely plateau6h,12h,24h and48h group were significantly higher than that in the plain control group (P<0.01). Conclusion1. The intestine may damage differently after the model rats acutely stressed to plateau, and the damage aggravate gradually with the exposure time extension of the acutely stressed to plateau.2. The expression of autophagy increased after the intestinal injury in model rats acutely stressed to plateau.
