Background;dental caries and periodontal diseases is chronic destruction of hard and soft tissues within oral cavity by a variety of microorganisms which is vary common diseases in humanity. The diseases forms through a complex interaction over time between acid producing bacteria and carbohydrate and too many host factors including teeth and salvia.oral microbial considered to be have an important role of caries and periodontal diseases etiology and pathogenesis, but precise role of microbial community structure and ecological complex remains to be fully established.Objective;Epidemiological investigation showed that characteristic in gansu ethnic minority yugu minority groups dental caries and periodontal disease prevalence is higher than other minority, this research used16srRNA gene clone library identification and analysis of yugu minority groups periodontal disease and dental decay differences in microbial community structure.Method;Yugu ethnic minority caries and periodontal disease rinse method to take saliva samples were used to establish16SrRNAgene cloning to obtain saliva total microbial DNA,using universal primers16SrRNA, recycling,ligation,transformation,build16SrRNA clone libraries and screening, NCBI BLAST database sequence comparison to identify a known sequence similarity.Phylogenetic tree was build by employing with CExpress, Blast, Clustal, Treeview, seaview, Mega, Bioedit, mothur software.Results;Clone library sequences in the NCBI BLAST database sequence comparison of dental caries in20clones and periodontal disease80cloned sequences. By using The mothur software were sequence analysis divided into4OTU analysis of the clone library sequences of dental caries. Library coverage value is90%, shanon-winner index, diversity index1.56.Through the mothur software for periodontal disease clone library of80clones were sequence analysis is divided into18OTU, library coverage value is76%, shanon-winner index, diversity index2.42.Conclusion;Use16SrRNA clone library analysis of microbial diversity Yugu periodontal disease and caries.Through the establishment of the proposed phylogenetic tree analysis phylogenetic relationship count out19kinds of dental caries and periodontal disease24kinds of strains of the species.This study analyzed samples of microbial diseases do not only control the difference between health and disease.Due tot he small sample size community richness value less than ideal,with More samples are required to explore the heterogeneity of periodontal microbial communities within individuals.
