Objective: To investigate the different expression and activation of Caspase-3isaffected the progress of myocardial apoptosis in some degree during the thecardiopulmonary bypass beating heart surgery and cardiac arrest surgery,providing new ideas for clinical work.Methods:Clean and healthy rabbits were randomly divided into two groups (n=12), the experimental group(the cardiac arrest group)and the control group(thebeating heart group). The control group established CPB model via routinecanulas, and the ascending aorta was clamped when temperature was reduced to28-30℃, the4℃st.Thomas ll cardioplegic solution was infused into ascendingaorta(15ml/kg).Using half of the first dosage of cardioplegic solution to infuseagain after15min and40min. Rewarming was after the third infusion andreperfusion was began at34℃.making a parallel bypass while the heart re-beat.Stop the cardiopulmonary bypass when the vital signs.stabled. The experimentalgroup established CPB model via routine canulas without clamped the ascendingaorta and reduced the temperature.Slowly stopped the bypass after120minutes..the rabbits2hours alive after the operation were regarded as thesuccessful experiment. Myocardial samples were taken quickly. Q-PCR was measured the Caspase-3mRNA expression levels of myocardial mitochondrialand TUNEL method was measured myocardial apoptosis.Results:(1)Apoptosis index:The experimental group (4.24±1.39); the controlgroup(11.26±2.28).(2):The experimental group relative mRNA Caspase-3expression levels(0.434±0.024) was lower than the control group (1.058±0.081).Conclusion:(1)Cardiac arrest group Caspase-3mRNA are highly expressedthan the beating heart group.It indicated that Caspase-3is involved in theapoptosis of myocardial cells while beating heart cardiac surgery style providesa certain extent of Myocardial protection.(2)The apoptosis was observed in both groups.the apoptosis index of cardiacarrest group were higher than the beating heart group.
