objective: to study the matrine for in vitro culture system of hyperplasticscar fibroblasts proliferation inhibition and the expression of MMP-1, MMP-9,to explore the mechanism of action of matrine inhibit hyperplasia scar, as toprovide theoretical basis for its application in clinical.Methods:10cases of resection of hyperplastic scar by burn patients(all insix months to one year after burns), the tissue block method for primary cultureof hyperplastic scar fibroblasts, take3-7generations in logarithmic growthphase cells in this experiment.The different concentration of matrine (0mmol/l,2.5mmol/l,5mmol/l,10mmol/l)of matrine respectively applied to the above inlogarithmic growth phase of the relative stability of hyperplastic scar fibroblasts,drug effect after24h,48h, cell proliferation inhibition rate as the index, with adetermined by MTT (methyl azo thiazole blue) colorimetric determination ofdifferent concentrations of matrine of hyperplastic scar fibroblastsproliferation;By Enzyme linked immuno sorbent assay,(ELISA) kits to join thedifferent concentrations of matrine effect after24h,48h, cells in supernatant ofmatrix metalloproteinases (MMP1, MMP-9) expression level of influence. Results:(1) of different concentration of matrine on human hyperplasticscar fibroblasts in vitro culture of growth has obviously inhibitory effect (p <0.05), and presents the concentration dependence and time dependence, ofwhich10tendency for strongest inhibition rate in group of10mmol/l,24h and48h after drug inhibition rate respectively was34.9%,63.6%.(2) theconcentration of matrine on the in vitro culture system of fibroblasts after24hand48h, the expression of MMP1and MMP-9quantity increased comparedwith control subjects, the difference was statistically significant, p <0.05).Conclusion: matrine on human hyperplastic scar fibroblasts in vitro culturehas inhibitory effect, within the scope of the experiment show dosedependent.Within the scope of the different concentration of bitter and alkali canpromote Matrix metalloproteinases (MMP1, MMP-9) expression, able todegrade the Extracellular Matrix component (Extracellular Matrix, ECM).In thehyperplastic scar on the prevention and treatment has the very good applicationprospect.
