ObjectiveWe detect the expression of Caspase-9and Livin in primary duodenal adenocarcinomaand analyzed the relationship among them by immunohistochemical method. we hopeto know the relationship between their expression and clinic significance in primaryadenocarcinoma of the duodenum.MethodsCollected60cases of primary duodenal adenocarcinoma patients clinical data,Whiledeployed its archived paraffin wax block specimens and30cases of adjacent tissues.Weuse SP immunohistochemical method to detect the expression of Caspase-9and Livin inthat cases,and to analyze the relationship between the expression of them.By combiningclinical data to analyze the relationship between the expression of primary duodenaladenocarcinoma of Caspase-9and Livin and the patient’s pathologicalcharacteristics.All data were processed by SPSS17.0.Results(1)Expression of Caspase-9Caspase-9in the adjacent tissues and cancer tissues can bepositive.The positive expression rate of Caspase-9in primary duodenal adenocarcinoma was25.0%, which was significantly lower than in normal tissues samples,(χ2=21.855,P<0.01); The expression of Caspase-9in primary duodenal adenocarcinoma wereassociated with the differentiation and lymph node metastasis but had nothing to dowith age,sex, tumor size,initial occurring position. depth of infiltration.Caspase-9positive expression in poorly differentiated cancer type was much lower than inmoderately and well differentiated types (χ2=4.375,p=0.036); The expression rate inpositive lymph nodes was much lower than that of negative lymph nodes(χ2=4.375,p=0.011);(2)Expression of Livin Livin in the adjacent tissues and cancer tissues can bepositive.The positive expression rate of Livin protein in primary duodenaladenocarcinoma was71.7%,which was significantly higher than in normal tissuessamples,(χ2=16.493, P<0.01). The expression of Livin in primary duodenaladenocarcinoma were associated with the depth of infiltration and differentiation andlymph node metastasis but had nothing to do with the other clinical pathology.Livinpositive expression in invaded mucous membrane and muscle was significantly lowerthan invaded tunica externa and out of tunica externa(χ2=7.074,p=0.008). Livinpositive expression in poorly differentiated cancer type was significantly higher thanin moderately and well differentiated type(χ2=6.482,p=0.011); The expression rate inpositive lymph nodes was significantly higher than that of negative lymphnodes(χ2=9.175,p=0.002)(3)Expression correlation of Caspase-9and Livin In the15positive expression ofCaspase-9cancer cases, there were7cases that positive expression of Livin; In the45negative expression Caspase-9cancer cases, there were36cases that positiveexpression of Livin; Livin and Caspase-9protein expression in primary adenocarcinomaof the duodenum was negatively correlated.(P<0.05,r=-0.320). Conclusions(1) Caspase-9and Livin in primary duodenal adenocarcinoma and normal duodenaltissue were expressed.(2) Caspase-9has a very close relationship with the occurrence and development ofprimary duodenal adenocarcinoma. When the expression of Caspase-9inhibition maylead to the development of tumor and lymph node metastasis. Can reflect the extent andprogress of tumor malignancy.Can help to diagnose primary duodenal adenocarcinoma,and make a preliminary evaluation of the prognosis and treatment of patients.(3)Livin and the development of primary duodenal adenocarcinoma also has a veryclose relationship. When the increased expression of Livin promote tumor developmentand lymph node metastasis, It can play a role in the early diagnosis and prediction oftreatment of primary duodenal adenocarcinoma.(4)Livin and Caspase-9expression in primary adenocarcinoma of the duodenum wasnegatively correlated, there may be mutual inhibition between the two, to participate intumorigenesis and development.(5)Low expression of Caspase-9and overexpression of Livin are closely related to theformation and progression of primary duodenal adenocarcinoma, for the future ofprimary duodenal adenocarcinoma targeted biological therapy to provide new ideas.(6)The experiment makes up Caspase-9and Livin research in primary duodenaladenocarcinoma blank, improve gastrointestinal cancer research both in the system,more fully expressed between it and gastrointestinal tumors relationship.
