Background Idiopathic pulmonary fibrosis is a common diseasethat leading to respiratory failure. In IPF, it often creates the lungorganization elasticity to drop, the lung volume reduces, the lung functioncarrying on decreases. There is lack of drugs what are efficient in clinicalapplication. So searching and exploitation for a new drug that can treatIdiopathic pulmonary fibrosis are the destination of all researchers.AKF-PD is a novel pyridine compound which is synthesized by our ownstudy group. In our previous study, it has been proven that it can inhibitsignificantly the proliferation of fibroblasts. So we decided to carry outanimal trial to confirm the prevent effect of AKF-PD in pulmonaryfibrosis caused by BLM.Methods Forty eight institute of cancer research (ICR) micewere randomly subdivided into normal contrast group, AKF-PD treatedgroup, pulmonary fibrosis model group, and enalapril-treated group.The mice in the AKF-PD-treated group, model group, enalapril-treatedgroup underwent injection of bleomycin (10mg/kg.d) through venacaudalis. One day before the injection begins, AKF-PD group tookAKF-PD(500mg/kg.d), enalapril group took enalapril(10mg/kg.d),normol and model group took the dissolvent of AKF-PD at a samevolume as AKF-PD and enalapril. After 28 days of the experiment, all animals were sacrificed and the lungs were collected for HE and Massonstaining, immunohistochemical staining and western-blotting for collagenⅠ.Results 28 days after treatment, in model group pulmonarycoefficient and pulmonary damage index of the lung are higher than thenormal contrast group (p<0.01). by contrast with model group, inAKF-PD treated group, lower index is found (p<0.01), and inenalaprol-treated group, no significant change is found (p>0.05). Thesemi-quantity determination of collagen I in model group increase(p<0.01). by contrast with model group, in AKF-PD treated group, thiscontent decreased (p<0.01), in enalapril group, no significant change isfound (p>0.05).Conclusions These results demonstrated that AKF-PD canprevent pulmonary fibrosis significantly caused by bleomycin in mice.
