Experiment 1 The study on the function of analgesia of the total lactones of ginkgoObjective: To know the effect on the funtion of analgesia of the total lactones of ginkgo.Method: The pain mice models caused by hot stimulus and acetic acid were applied to testthe pharmacodynamics. Result: The mice’s threshold of pain to hot stimulate and chemicalstimulus all can be raised by the three different dosages of total ginkgolides which are 2.5,5and 10 mg/kg. There are significant difference between high dosage and blank controlgroup (P<0.05, 0.01). Conclusion: The total ginkgolides has obvious effect of analgesia torheumatoid arthritis.Experiment 2 The study on the function of anti-inflammation of the total lactones ofginkgoObjective: To know the effect on the function of anti-inflammation of the total lactones ofginkgo Method: Test the pharmacodynamics which were the increase of abdominal cavitycapillary permeability in mice caused by acetic acid, auricle swelling induced by croton oilin mice and granuloma induced by the plastics wreath in mice. Result: Compared withblank control group, the high dosage of total ginkgolides has lower belly cavity liquid ODvalue, and the differences are significant. Compared with blank control group, all thedosage of total ginkgolides decrease the swelling of the inflammation ear. The wet weightand dry weight of granuloma in high and middle dosages are less than those in blankcontrol group, and the differences are significant (P<0.05,P<0.01). Conclusion: The totalginkgolides has some function of anti-inflammation.Experiment 3 The effects of the treatment of the total lactones of ginkgo against CIAmiceObjective: To know the effects of the treatment of the total lactones of ginkgo against CIAmice.Method:By using typeⅡcollagen induced arthritis model in mice,observe its bodyweight,erythema,tail node and the feet swelling degrees in mice and make pathologicalsection and grade arthral lesion.Result: 2.5,5 and 10mg/kg of the total lactones can increasethe weight of CIA mice.(P<0.05)and relieve the incidence of arthritis .It can also inhibit thefoot swelling and swelling scoring significantly. Histopathologicdl examination and resultof the degree of tissue deterioration showed that it can improve the inflammatory cells,synovium hyperplasia and the normal structure of joint .the degree of tissue deterioration of the high, middle dose of ginkgolides was smaller than that of the modelgroup. Conclusion:The total ginkgolides can cure the mutlisymptom of rheumatoid arthritisof CIA mice.Experiment 4 The mechanism of the total laetones of ginkgo against CIA miceObjective:To know the The mechanism of the total lactones of ginkgo against CIAmice.Method: Observe the stimulation index of rheumatoid arthritis mouse splenocytesunder the conditionsof ConA and LPS by using CIA mice model .DeterminateIL—2,IFN—r, NO induced from spleen lymphocyte and IL-1βand TNF-αfrom the culturesupernatant of CIA mice’s synovial cells.Result(1).The stimulation index of rheumatoidarthritis mouse splenocytes under the conditions of ConA and PLS was bigger than that ofnormal group. Total ginkgolides can inhibit the multiplication of T cell and B cell. (2)Innormal 1640 culture medium, the level of IL—2 had no significant difference between themodel and normal group. But the result was reversed once in the effect of ConA.Withstimulation of ConA, the level of IL—2 of the high, middle dose of ginkgolide was lowerthan model group and had significant difference.(3) In normal 1640 culture medium, thelevel of IFN—r all had no significant difference between every treatment group and normalgroup. The level of IFN—r increased in the stimulation of LPS and ConA.Compared withnormal group, there was significant difference. The level of IFN—r of the high, middledose of ginkgolides was lower than model group and had significant difference. (4)Innormal 1640 culture medium, the level of NO induced by every group’s spleen lymphocytehad no significant difference. With stimulation of ConA, the level of NO induced by modelgroup increased and had significant difference with the normal group. NO level of the high,middle, low dose of ginkgolides was all lower than model group and had significantdifference. Every group mice’s spleen lymphocyte had no significant difference in excretingNO by stimulation of LPS. (5)Content of IL-1βand TNF-αfrom the culture supernatant ofCIA mice’s synovial cells in model group was all higher than the normal group. They hadsignificant difference. Content of IL-1βand TNF-αfrom the culture supernatant of CIAmice’s synovial cells in ginkgolides treatment group was lower than the model group. Theyhad significant difference. Conclusion: The mechanism of total ginkgolides’s inhibition ofswelling of the rheumatoid arthritis have some relation with inhibition on themultiplication of T cell and B cell and the inhibition on the prodution of IL-2 ,IFN-r andNO.
