Object Duchesnea indica(Andr.)Focke had much abundant germplasm resources,which could used by gardening and medicinal.Moreover, Duchesne aindica(Andr.)Focke widely distribute,had strong adaptability and high developing value. Currently,the research mainly focused on the chemical composition and pharmacological effects,using molecular markers to explore the germplasm genetic diversity had not been reported.The24samples of different regions in Hunan province were analyzed by RAPD molecular markers in this study,the results not only provide a reliable theoretical basis for making use of local Duchesnea indica(Andr.)Focke biotype but also have the significant to the future breeding work of Hunan province.Methods The univariate and orthogonal design were used to optimize PCR reaction system.Using PopGene32software to calculate the parameters of genetic diversity between all the samples,using the UPGMA in the NTSYS programs to analyze the genie relationship of every samples.Results l.The suitable RAPD reaction system for Duchesnea indica (Andr.)Focke genomic DNA was:20μl RAPD reaction volume containing1.5mmol/LMg2+,250μmol/LdNTPs,lUTaqDNApolymerase,0.2 μmol/L primer,60ng templates DNA;2.The analysis of RAPD marker showed that14RAPD primers amplified a total of90scorable markers,percentage of polymorphic loci was90%,Nei’s genetic diversity and shannon’s information index were0.3128and0.4675.The results of RAPD showed that the genetic diversity of Duchesnea indica(Andr.)Focke in Hunan was rich.3.The cluster analysis indicated that24samples were classified into two group in the threshold of0.52distance:Changde NO1,NO2,PingjiangNO2and Lixian were a large class,the rest were the second largest category. The relationship of ChangdeNO2and Botanical garden were far.
