Herbal as traditional medicines, which has little toxicity and adverse effects,has an important role in modern drug research and development. Coltsfoot Flower asa traditional medicine, has the effect of the lungs cough, phlegm and asthma, the maincomponent containing more, mainly produced in Shanxi, Henan, Shanxi Guanglingnational of Chinese medicine the Coltsfoot of GAP planting base., Coltsfoot as thefocus of attention, with good prospects.Capillary electrophoresis, as a new chromatography technology, is a widelyapplied technique in separation science on account of its high efficiency, rapid rate ofseparation and small sample requirement and has offered satisfactory results in theanalysis of some Chinese herbs.In this paper, high performance capillary electrophoresis(HPCE) was used toresearch qualitative and quantitative of Flos Farfarae, which provided the basis forthe quality control of Flos Farfarae.In chapter1: The genereal situation of the basic principles and research progressof HPCE, and it’s application in the main active ingredient of Chinese medicine andtraditional Chinese medicine preparation. The genereal situation of the study oftraditional Chinese medicine Coltsfoot description, including the study of thechemical composition, pharmacological activity, toxicological, clinical applicationand quality control.In chapter2: To develop reverse flow non-aqueous capillary electrophoresis(RF-NACE) method for determination of tussilagone in Flos Farfarae anddetermination completed within5minutes. The optimal electrophoresis conditions:Electrophoresis was performed on a75μm×40.2cm (effective length is10.2cm)fused silica capillary,20mmol/L Sodium acetate in methanol as the backgroundelectrolyte.The running voltage was30kV,the detection wavelength was at214nmand the temperature was25℃. injection conditions:0.5psi,3s. Determination ofdifferent origin and different businesses tussilagone in Flos Farfarae, is a simple,low-cost,effective and reliable method. In chapter3: To develop high performance capillary electrophoresis (HPCE)fingerprints of Flos Farfarae and show26characteristic common peaks, amongwhich6peaks were recognized. Electrophoresis was performed on a75μm×57cm(effective length is50cm) fused silica capillary.30mmol/L sodium borate contained25%methanol adjusted to pH8.8was applied as the background electrolyte. Therunning voltage was20kV, the detection wavelength was at214nm and thetemperature was25℃,the injector conditions:0.5psi5s. The control fingerprint ofHPCE of Flos Farfarae was established, and the similarity of fingerprintchromatogram of Flos Farfarae from different places were compared. It has guidingsignificance of quality control of Flos Farfarae.
