Blumea balsamifera DC. is a folk herbal medicine, mainly in Yunnan, Guizhou, Guangdong, guangxi and Taiwan province. Whole plant or aerial parts used as medicine, which is treat for analgesic,diaphoretic, expectorant cough etc. It is widely used in Yi, Miao, Zhuang, Li nationalities with a long history, especially used as a treatment for lung cancer and has significant curative effect in the Yi nationality living area. Its methanol extraction has obvious inhibitory effect on several kinds of tumor cells.The medicinal materials was extracted with95%methanol, and methanol extract was supended with water and fractionated with same volume of petroleum ether, methylene chloride, n-butyl alcohol. The active fractions were isolated by silica gel, sephadex LH-20, reverse phase C18column chromatography, fast preparation and semi-preparation HPLC, and their chemical structures were identified with NMR, Mass and UV spectrum. The antitumor activity for different fractions and part compounds were assayed by MTT methods using human lung carcinoma A549cells in vitro.Nine compounds were isolated From two fractions and eight compounds were identified as liquiritigenin (1), p-sitosterol (2), kuma-takenin (3), glycyrol (4), isoglycyrol (5), focosyl caffeate (7), inuch-inenolide B(8), neogaillardin (9).The glycyro(3), iso-glycyrol(5), inuchinenolide B (8) and neogaillardin (9) were isolated from this herb for the first time.From the cell experience, the results indicated that methylene chloride fraction and petroleum ether fraction have a strong inhibition on the proliferation of A549cells, followed by n-butyl alcohol fraction and water fraction. Compounds1,3,5,7have a significant inhibitory activity on the proliferation of A549cells and the IC50were11.52μg/ml,20.69μg/ml,7.74μg/ml and5.65μ.g/ml, lower than the IC50(21.03μg/ml) of Rg3.The inhibitory activity of Compounds1,3and5increases depending on drug concentration increase. Compound7showed a high inhibitory activity in low doses, and presents no obvious dose-dependent. Compound8and compound9also showed a weak inhibitory activity, with a dose-dependent relationship.This article study on the different fractions and compounds from Blumea balsamifera DC.about the inhibition for the proliferation of A549cells, it provides a method for the treatment of lung cancer and other turmors.
