Aim:Through CLSM,we observe the inhibitory effect of Magnolol act on Streptococcusmutans biofilm in vitro,and preliminary study caries virulence factors of Magnolol act on Str-eptococcus mutans biological film:acid production,acid resistance, exopolysaccharide forma-tion biological film,transcription expression effect of formation ability,in order to discuss Mag-nolol Pharmacological action mechanism.of preventing cavities.Method: First of all,we establish vitro model of Streptococcus mutans biological film onthe coverslip, we each put AO/EB dyeing in it after different drug concentration act to observeand conduct red green fluorescence quantitative analysis by using CLSM: According to GenBank gene pool,we inquire gene sequence: ffh、gtfD、pdp etc,and design primer to measurethe expression level of the causing caries virulence factors ffh、gtfD、pdp gene fragment ofStreptococcus mutans biological film which is acted by different drug concentration by RT-PCR.Result:The green fluorescence area of the experimental group is decreased following thedrug concentration increase,each drug concentration all obviously reduces the growth activityof Bacterial biofilm than the control group, viable ratio is decreased obviously inside film afterobserving Magnolol acted on bacterial biofilm,which is formed24hours by CLSM.In the caseof gradient drug concentration is ensured the consistent of amount of template by the electrop-horesis gel imaging system to measure and analyse, the Brightness of virulence factors:ffh,gtfD,pdp,etc.gene fragment become dark,even disapeared on the band of electrophoresis detection,which indicate: Magnolol make the virulence factors expression quantity of Streptococcusmutans biofilms obviously reduce trend with a certain concentration.Conclusion: Magnolol can restrain well the growth activity of Streptococcus mutans bio-films,and suppress transcriptional expression of mRNA of the causing caries virulence factors:ffh,gtfD,pdp,etc.gene fragments,Thus we Infer that Magnolol has effect on the growth of Strep-tococcus mutans biofilms is due to restrain or disturb transcriptional expression of some genefragments,so as to lead to the struction of biofilms be damaged.
