ObjectiveObservation of Yangwei Jiedu Decoction on sepsis model in rats, to explorethe stomach Jiedu Decoction for the treatment of sepsis mechanismMaterials and methods60SD rats were randomly divided into four groups, respectively, for thesham operation group (group A), sepsis (CLP) model group (group B), YangweiJiedu Decoction treatment group (group C), control group (group D) ofamoxicillin. Using a cecal ligation and perforation method for B, C, D threegroups of rats were sepsis model replication, the rats in group A operation withmore than three groups, but without perforation. After oral administration of8h a corresponding drug intervention, group B received saline by oral gavage,continuous dosing6times. In the experimental process of rats were observedin general status, mortality rate. At the end of experiment observation ofcolonic tissue pathological changes, and the use of enzyme-linked immunosorbentassay (ELISA) method on serum endotoxin (endotoxin), tumor necrosisfactorα(TNF-α), IFN-γ(INF-γ) was detected. Measurement data wereconfirmed by SPSS13.0software to carry on statistical analysis.Results1false operation group rat mortality was1%; sepsis model rat mortalitywas46.7%; Yangwei Jiedu Decoction group rat mortality was26.7%; amoxicillinrat mortality was40%.2in serum of model rats with endotoxin, TNF-α, INF-γwas significantlyhigher than that in sham operation group (P <0.01); stomach Jiedu Decoctiongroup in serum of rats with endotoxin, TNF-α, INF-γlevels were significantlylower in sepsis model group (P <0.01); amoxicillin controls in the blood serum of rats endotoxin in comparison with the model group had no significantdifference, TNF-α, INF-γwas significantly lower than that of sepsis modelgroup (P <0.05).3false operation group rats colon tissues showed no significantpathological changes of inflammatory cells, occasionally appearing. A model ofsepsis rats colon tissue microscopically the rats colon tissue oedema,infiltration of inflammatory cells, epithelial cell necrosis. Stomach JieduDecoction group rat colonic histopathology microscopically the infiltration ofinflammatory cells, edema is greatly reduced compared with the model group, noepithelial cell shedding phenomenon. Amoxicillin rats colon tissue slices wereobserved under microscope and inflammatory cell infiltration, slightly less thanthe stomach Jiedu Decoction group, the degree of edema was significantly lighterthan that in the model control group.Conclusion:1stomach Jiedu Decoction can obviously reduce the sepsis model of ratcolonic tissue inflammatory reaction.2stomach Jiedu Decoction can significantly reduce sepsis model in rats serumendotoxin, tumor necrosis factorα and IFN-γcontent.3stomach Jiedu Decoction on sepsis model rats and its therapeutic effectmay be through its reducing the model rats serum tumor necrosis factorα andIFN-γcontent and realization.
