Fructus forsythia is one of important traditional medicine plant in China. In this study, an experiment was designed to study the optimization of the extraction of the active ingredients in this plant by response surface methodology (RSM). Three independent variables (extraction time, extraction times, ratio of water to sample) were optimized. The anti-inflammatory activity of the active components was studied. The pharmacokinetics of phillygenin was investigated at the same time. The results showed that the best ratio of extraction was13.577mg/kg and the optimized conditions were extraction time34.6min, ratio of water to sample22.3mL/g, and three extraction cycle. The anti-inflammatory experiment indicated that phillygenin had the best activity. The anti-inflammatory activity of phillygenin was dose dependent. The pharmacokinetic parameters of phillygenin in rat were determined by HPLC. The main parameters of phillygenin in rat plasma were as follows:t1/2z were6.02,5.62and5.79min separately. Phillygenin was distributed and eliminated quickly.
