Purpose:To discuss the effect of Taohong Siwu Decoction regulating endothelial cellfunction and treating arteriosclerosis obliterans (ASO).Material and method:Material:6~8month old Wistar rats of clean grade50, weight (200±20) g, provided byexperimental animal center of Liaoning University of Traditional Chinese Medicine.Composition of high fat diet:62.8%basic feed+20%lard+150g/L cholesterol+20g/Lsodium cholate+2g/L propylthiouracil. Enzyme linked immunosorbent assay (ELISA)double antibody sandwich detection kit were purchased from Shanghai Yueyan biologicaltechnology company, Interleukin-1(IL-1, batch number AE98022Ra), Tumor necrosisfactor-α (TNF-α, batch number AE90133Ra), Endothelin-1(ET-1, batch numberAE90650Ra). Nitric oxide (NO) detection kit was purchased from Nanjing Jianchengbiological technology company. Rabbit anti mouse anti factor Ⅷ-fluorescein isothiocyanate(FITC) antibody was purchased from Wuhan boster company.Methods:ASO models were constructed with the method of high-fat diet plus intimal injury,the rats were rando毫升y divided into model group, Taohong Siwu Decoction low, medium,and high concentration group, each group had9-12rats. Another10Wistar rats were chosenas normal group with clean level.8weeks after building model, limb blood flow perfusionwere observed using laser doppler blood flow meter, light microscope and transmissionelectron microscope artery gross morphology, the number of circulating endothelial cells(CECs) were deter分'ed using Percoll density gradient centrifugation. Serum tumornecrosis factor alpha (TNF-α), interleukin1(IL-1), endothelin1(ET-1) and nitric oxide (NO)level were detected using enzyme-linked immunosorbent (ELISA) double antibody clipmethod.Results:ASO model was established successfully. Compared with normal group, afterbuilding model,the left hind leg blood flow perfusion group rats were decreased. Comparedwith model group, the blood flow perfusion improved significantly in the peach siwu decoction group medium, high concentration group (P <0.01). Compared with the normalgroup, serum cholesterol, triglyceride, low density lipoprotein cholesterol (HDL-C) levelobviously increased, lower high-density lipoprotein cholesterol (HDL-C) level significantlydecreased in model group of rats, there were significant difference (P <0.01). Compared withmodel group, serum cholesterol, triglyceride level declined obviously in Taohong SiwuDecoction high concentration group, there was significant difference (P <0.01). Normalgroup had few quantity of CECs in the peripheral blood, CECs volume increased significantlyin peripheral blood of rats in building groups, compared with normal group, the differencewas significantly (P <0.01). Compared with model group, CECs quantity in Taohong SiwuDecoction high concentration group was obviously decreased. Serum TNF-α and ET-1, IL-1significantly elevated in model group compared with the normal group, serum NO leveldecreased significantly than the normal group, the differences were significant (P <0.01),after the application of decoction containing four drugs of persicae and carthami treatment,serum TNF-α and ET-1, IL-1, NO level of each group also had improved, the change wasmost obvious in the high concentration group.Conclusion:1. About the mechanism of ASOThe injury and abnormal function of endothelial cells is the important pathological process ofASO.2. Mechanism of Taohong Siwu Decoction in the treatment of ASOTaohong Siwu Decoction could protect vascular endothelial cell, improve function ofvascular endothelial cells, and contribute to the treatment of ASO.
