Objective:To investigate the therapeutic effect of Different compatibilitysaltpetre and alum pulvis on alpha-naphthylisothiocyanare(ANIT) inducedintrahepatic cholestasis in rats.Methods:Using ANIT induced intrahepatic cholestasis rats model70, wererandomly divided into model group, western medicine and traditional Chinesemedicine (TCM)1-4group, in addition to the model group and20, the rest ofthe each group of10, only made mould and to choose not only as a normal controlgroup10rats. Group all day a lavage, model group and normal control group at6ml/kg shall be0.9%sodium chloride injection and western medicine groupaccording to0.1g/kg/d for mattel, traditional Chinese medicine (TCM)1-4groupsaccording to the2ml/d respectively for potassium nitrate add alum, glauber’ssalt, add alum, potassium nitrate and copperas, glauber’s salt and copperas asmain compatibility of saltpeter aluminite powder to fill the stomach. Drugdelivery time starts from building the day, after48h execution model group,10rats serum standby to return.7days to be put to death all the animals.Fullautomatic biochemical analyzer tested indexes of liver function (TBIL、DBIL、TBA、ALT、AST、ALP)in each group,enzyme-linked immunosorbent kits to determinethe Na~+-K~+-ATPase activity, the polymerase chain reaction tested The expressionof Anion exchange protein2(AE2,Anion Exchanger2)mRNA in rats, observedpathological changes in liver tissues under the light microscope,andtransmission electron microscopy to observe the ultrastructure of liver cells.Results:(1)Intrahepatic cholestasis, SD rats serum total bilirubin (TBIL),direct bilirubin (DBIL), alanine aminotransferase (ALT), alkaline phosphatase(ALP) and total bile acid (associates) levels were significantly elevated (P<0.01). Compared with48h after building, the experiment is finished modelgroup serum TBIL, DBIL, associates levels were significantly decreased (P <0.01). When the experiment is complete, compared with model group, western medicineand traditional Chinese medicine (TCM)1-4groups of rats serum TBIL, DBIL,associates were significantly reduced (P <0.01), Chinese medicine group1(potassium nitrate, alum) rats serum TBIL, DBIL, associates level did not seeobvious difference compared with the western medicine group (P>0.05),Withtraditional Chinese medicine (TCM)2(potassium nitrate, copperas group),traditional Chinese medicine (TCM), three groups (glauber’s salt, alum), fourgroups of Chinese medicine group (glauber’s salt, copperas) compared to thedifference was statistically significant (P <0.05). Compared with normalcontrol group, western medicine group, Chinese medicine1-4ALT activity hasno obvious difference (P>0.05), Compared with model group, the treatment groupwere significantly different (P <0.01). Compared with model group, westernmedicine and traditional Chinese medicine (TCM)2-4group ALP activity was nosignificant difference (P>0.05), Traditional Chinese medicine (TCM)1set ofALP activity compared with the model group had significant difference (P <0.01).Each aspertate aminotransferase (AST) in serum activity of no significantdifference (P>0.05). Compared with model group,2group of western medicineand Chinese medicine treatment group1-4Na+-k+atpase activity wassignificantly increased (P <0.01),1Chinese medicine group and western medicinegroup compared with no significant difference (P>0.05), Chinese medicine group1and traditional Chinese medicine (TCM)2-4compared to group the differencewas statistically significant (P <0.05).(3) intrahepatic cholestasis, AE2mRNAexpression of the model group increased significantly (P <0.01), westernmedicine and traditional Chinese medicine (TCM)1-4group all can reduce itsexpression, compared with the model group had significant difference (P <0.01),Chinese medicine group1compared with western medicine and traditional Chinesemedicine (TCM)2-4group the difference was statistically significant (P <0.05),but the treatment group AE2mRNA expression were still significantly higher thanthe normal group (P <0.01).(4) then visible model group structure of hepaticlobule is incomplete, the outline is not clear, have scattered point necrosis; Bile duct epithelial cell swelling, necrosis, lumen wide; Portal area has a largenumber of inflammatory cells infiltration. Western medicine group, hepaticlobule structure is relatively complete, only individual liver cell in vacuolesdegeneration. Compared to2-4Chinese medicine group, Chinese medicine group1(potassium nitrate alum) hepatic lobule structure more complete, roughlynormal interlobular bile duct, portal area only a small amount of inflammatorycell infiltration, hepatic cell degeneration, necrosis, to a lesser degree.5]visible model group under the electron microscope mitochondria swelling in thecytoplasm, some mitochondria cristae broken, show cavitation model change; Doesnot change significantly smooth endoplasmic reticulum, glycogen is more, golgibody hypertrophy; Nucleus edge gathered, and the larger nucleus, intranuclearheterochromatin increased; Hepatic blood sinus in Kupffer cells, irregular shape,cytoplasm contains a large amount of mitochondria and lysosome; Part of livercell organelles to dissolve; Capillary bile duct expansion to different extent,and decrease microvilli in the capillary bile duct, bile duct around the denseband broadening or outer layer. The dissolution of western medicine group ofcytoplasm, nucleus pycnosis, contraction mitochondria, smooth endoplasmicreticulum expansion, rough endoplasmic reticulum.1-4group in Chinese medicine,Chinese medicine group1(potassium nitrate alum) is most close to normal livercell structure, cell organelle is rich, have plenty of glycogen granules.Conclusion:The saltpetre and alum pulvis with Potassium nitrate, alum asthe main compatibility can improve of intrahepatic cholestasis when abnormalserum biochemical indicators,and enhance the Na~+-K~+-atpase activity. Promotingblood qushi method can improve abnormal serum biochemical indicators at the timeof intrahepatic cholestasis, raised the level of LP-X, increased expression ofNa+-H+exchanger mRNA, significantly reduced the liver cell degeneration,necrosis and capillary bile duct dilatation, for ANIT induced intrahepaticcholestasis in rats have a significant therapeutic effect. Reduce AE2mRNAexpression, reduce liver cell degeneration, necrosis and capillary bile duct expansion, for ANIT intrahepatic cholestasis rats caused by a significanttherapeutic effect.
