This thesis contains three parts, namely the survey studies on the Syringa, the research on the chemical constituents of Syringae Cortex and the developing of a HPLC method for the simultaneous determination of syringin and oleuropein in Syringae Cortex.The first part of the thesis is the survey studies on the Syringa through summarizing the chemical constituents, pharmacological action, quality control of the plants mainly. The Syringa contains phenylpropanoids, iridoid and secoirdoid components, phenethyl alcohol, triterpene and steroidal components, flavonoid and simple organic compound mainly; Pharmacological actions are about anti-microbial, anti-inflammatory, antioxidant, protect liver and cholagogue, relieve cough and eliminate phlegm, anti-virus and so on; High performance liquid chromatography, gas chromatography, gas chromatography-mass spectrometry and ultraviolet-visible spectrophotometry are used usually to control the quality.The second part of the thesis is the research on the chemical constituents of Syringae Cortex including the extraction, separation and structural identification of the chemical constituents and the component analysis of Syringae Cortex by ultra-performance liquid chromatography with quadruple-time-of-flight mass spectrometry (UPLC-Q-TOF-MS).By using a variety of chromatographic techniques and spectroscopic methods,3pure compounds have been successfully isolated from the75%ethanol extract of the Syringae Cortex, including one phenylpropanoids. syringing (I) and two iridoids. they are oleoside-11-methyl ester (Ⅱ) and oleuropein (Ⅲ). oleoside-11-methyl ester was isolated from this plant for the first time. By using UPLC-Q-TOF-MS, Scifinder data base, ChemicalBook data base, Chemspider data base, and the references,17proposed compounds have been found from the methanol extract of the Syringae Cortex in positive and negative ion mode respectively, of which8proposed compounds were found from this plant for the first time. This study identified the chemical compounds from the methanol extract of the Syringae Cortex by UPLC-Q-TOF-MS firstly, supplied the evidence to the material foundation of pharmacodynamic action of Syringae Cortex.The third part of the thesis is the developing of an HPLC method for the simultaneous determination of syringin and oleuropein in Syringae Cortex, improving the quantitative determination of Syringae Cortex in Pharmacopoeia of the PRC2010, Volume I.12batches of Syringae Cortex were determinated. The yields of the two components in different batches exhibit significant differences. This method is simple, quick and repeatable, improves the specificity of the existing, which can be a good method for the quality control of Syringae Cortex.
