Objective UPLC-Q-TOF/MS was used to investigate the chronic superficial gastritis, early gastric cancer and advanced gastric cancer’s plasma metabolomic characteristics. By this method, we seek the potential early diagnosis biomarkers of gastric cancer, and discuss the metabolism molecular events which early gastric cancer development may involve.Methods This study applied UPLC-Q-TOF/MS to test metabolomic characteristics in the plasma, which contained fifty cases of chronic superficial gastritis(CSG), seven cases of early gastric cancer(EGC) and twenty-five cases of advanced gastric cancer(AGC). The plasma was collected from the Yangzhong city, Jiangsu Province, which is a high incidence area of gastric cancer. Then, using principal components analysis (PCA), partial least squares-discriminant analysis (PLS-DA) and Orthogonal partial least squares-discriminant analysis(OPLS-DA) to determine the potential tumor markers in early diagnosis of gastric cancer and also discuss the metabolism molecular events which early gastric cancer development may involve by analysing the variation characteristics of metabolomics above the three groups.Results The two and three dimensions score plot of OPLS-DA can effectively differentiate chronic superficial gastritis and early gastric cancer, early gastric cancer and advanced gastric cancer. Finally, we find that L-Carnitine, L-Proline, Pyruvaldehyde, PC(14:0/18:0), LysoPC(14:0), Lysinoalanine can distinguish early gastric cancer from chronic superficial gastritis. By comparing the early gastric cancer and advanced gastric cancer, we find that the changes of L-Proline, L-Valine, Adrenic acid, PC(O-18:0/0:0), LysoPC(20:4(5Z,8Z,11Z,14Z)), Pyruvaldehyde between the two groups have statistical significance.Conclusion UPLC-Q-TOF/MS is a rapid, sensitive and non-invasive technique of identifying biomarkers. The metabolic profile of chronic superficial gastritis and early gastric cancer, early gastric cancer and advanced gastric cancer are significantly difference. So the differences metabolites may become the potential early diagnosis biomarkers of gastric cancer.
