| This dissertation include three partsPart one:Literature reviewBased on the literatures related to Lawsonia inermis L. and Impatiens balsamina L., a review about chemical constituents, pharmacological activities and applications of these two plants were presented.Part two:Experimentation1TLC identification method for Lawsonia inermis L. and Impatiens balsamina L.The TLC identification method for Lawsonia inermis L. and Impatiens balsamina L. has been established according to the analysis of6batches of Lawsonia inermis L. and2batches of Impatiens balsamina L. The method is:Silica gel TLC plate, developing with chloroform-methanol(12:1) after saturated30min, monitoring under UV light of365nm. This method is fast and accurate for the identification of Lawsonia inermis L. and Impatiens balsamina L.2Study of HPLC fingerprintEstablished the HPLC fingerprint spectrum determination method of Lawsonia inermis L and Impatiens balsamina L. for the first time. HPLC fingerprint spectrum of6batches Lawsonia inermis L. and2batches Impatiens balsamina L were evaluated, which demonstrated that there is no significant chemical differences between Impatiens balsamina L. from different places, chemical differences between Lawsonia inermis L. from different places were observed, but still obvious common characteristics exist. In addition, when HPLC method employed to identify Lawsonia inermis L. and Impatiens balsamina L., determination can base on the polar constituents as well as the6characteristic chromatographic peaks.3Studies on the components of Lawsonia inermis LBase on literatures, chemical constituents library was established. UPLC/Q-TOF-MS method was established, which was employed to analysis chemical constituets of Lawsonia inermis L.. Major peak were identified according to retention time, molecular weight, molecular fragment peaks as well as the chemical constituents library of Lawsonia inermis L. The positive and negative ions was used to do the analysis,96peak were observed,17compounds and2compounds structure type were identified, including flavonoids, naphthoquinones, coumarins, glycosides and organic acids. The established method is simple and rapid for elucidation the constituents of Lawsonia inermis L. and laid the foundations for quality control of Lawsonia inermis L.4Studies on the characteristic components of Lawsonia inermis LEstablished the semi-synthetic method of lawsone monomethyl ether. Reflux was employed to do the semi-synthesis from lawsone, after reflux, sample washed with water and sodium hydroxide solution, and then purified to get97%lawsone monomethyl ether, which is simple and high efficiency. Established preparative liquid chromatography separating technology of1,2,4-trihydroxynaphthalene-l-O-β-D-glucopyranoside, luteolin-4’-O-β-glucoside and eugeniin in Lawsonia inermis L. The established method is simple and rapid and laid the foundations for quality control of Lawsonia inermis L.5Content of lawsone was evaluated by HPLCEstablished HPLC determination method of lawsone in henna. Regression equations of lawsone were y=50.386x-5.3621, r=0.9999, which showed that the calibration curve was linear within the range of0.50μg/mL-40.0μg/mL for lawsone. The method was simple, accurate, and with a good reproducibility. It provided references for further studies of it’s clinical pharmacology and security evaluation.Part three:summary and discussion1Established the TLC and HPLC identification method of Lawsonia inermis L. and Impatiens balsamina L for the first time.2Established the HPLC fingerprint spectrum determination method of Lawsonia inermis L and Impatiens balsamina L. for the first time.3Chemical constituents library of Lawsonia inermis L. and Impatiens balsamina L. was established for the first time.4Established the UPLC/Q-TOF-MSE qualitative method of the main components in effective part of Lawsonia inermis L. for the first time.5Established the semi-synthetic method of lawsone monomethyl ether for the first time.6Luteolin-4’-O-β-glucoside and eugeniin were separated from Lawsonia inermis L. for the first time.7Established the HPLC determination method of lawsone in Lawsonia inermis L. |
