Impatiens balsamina L.is dry herb which is belongs to Impatiens Balsaminaeeae L..And resources are very abundant.All parts of north and south China are cultivated.The medicinal value of the plant early cognitive by folk,Outside the brush on the sore or mash is used to treat fungal infections caused by fungal.Clinically used to treat rheumatism,paronychia,joint pain.The antifungal activity of Impatiens balsamina L.is value of scientific research,development and utilization.To optimize extraction technology of Impatiens balsamina L.by central composite design/response surface methodology in this paper.With ethanol concentration,extraction time and solvent ratio as independent variables,yield of total flavonoids from Impatiens balsamina L.as dependent variable,levels of independent variables were regressed by multiple linear and fitted by binomial,response surface method was used to optimize extraction process.Optimal extraction technology was as following:extracted 164min with 23 times and the amount of 81%ethanol.Deviation between observed and predicted values was-0.4%.The model of experiment was good predictability,simple,and good repeatability.To remove the impurity,macroporous resins was chosen to enrich and purify flavonoids from Impatiens balsamina L..With adsorbance and the recovery rate of total flavonoids as the criterion of evaluation for screening and process optimization.Optimum macroporous resin was AB-8,and the optimum condition was as follow:the sample solution was 9.415mg/ml resin,the concentration of sample solution was 1.3 8 7mg/ml(equivalent to 8.55mg/ml)with a flow rate of 5BV/h(1BV was 30ml),the eluant was 5BV water and 8BV 60%alcohol,elution flow rate was 4BV/h,the resin can be used repeatedly 8 times.The content of total flavonoids in the purified extracts reached 22.65%.The purity of total flavonoids increases 5.16 times than before.In vitro antibacterial activity of the extracts was studied on the Candida albicans and Microsporum canis by oxford cup method.The results showed that the extracts had clear antu-fungi activity on the two fungis,and the diameter on the Candida albicans was greater than the Microsporum canis.Using trace double dilution method recommended by CLSI,determination minimal inhibitory concentration(MIC)and minimal bactericidal concentration(MBC)of Impatiens balsamina L.total flavonoids extracts on different fungis,and MIC and MBC of Candida albicans were 0.7813,0.7813mg/ml,respectively.MIC and MBC of Microsporum eanis were 1.563,3.225mg/ml,respectively.It can be seen that the extracts had strong anti-fungi activity on the two fungis,which laied a foundation for the research of antibacterial activity of Impatiens balsamina L..
