Purpose:Study the therapeutic effects of Compound Yanshou Solution(YSY) in aged and hyperlipidemia rats and provide laboratory evidence for the research of reduce blood fat and slow aging of Compound yanshou solution.Method:60male SD rats were randomly divided into controlled group, model group, positive drug (fenofibrate) group, YSY group(low, medium and high doses group)of6groups based on their weight and10rats in each group. Control group rats were fed with standard diet and exposed to natural air circulation in toxicant cabinet for4h every day. SD rats of model group were fed withhigh-fat diet and exposed to ozone for4h (1.2±0.2ppm) every day. Positive (fenofibrate) group rats were oral administration YSY18ml/kg every day. YSY group rates were oral administration4.5,9,18ml/kg every day respectively. Each group rats were killed on d70.We detected the serum lipids, liver function, antioxidant enzymes and the function of related enzymes on d70;Study the pathological changes of heart, liver and brain via HE staining; immunohistochemistry showed that the content of the key oxidative stress enzyme in situ.All rats were killed on d70. Then we detected the content of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), lipid peroxidation (LPO), triglyceride (TG), total cholesterol (TC), high density lipoprotein (HDL-C) and low density lipoprotein (LDL-C). Pathology study:①Lung:Qualitative described the pathological change via HE staining, quantitative measured the alveolar volume difference and NOX2immunohistochemical expression, quantitative measured the collagen fibers collagen fibers ratio via masson staining.②Liver:Qualitative described the pathological change via HE staining and quantitative measured the hepatic cell edema and NOX2immunohistochemical expression, quantitative measured the collagen fibers collagen fibers ratio via masson staining.③Brain:HE and nissl staining qualitative described the pathological change, quantitative measured the number of hippocampal neuronal apoptosis, cortical neurons loss and the NOX2immunohistochemical expression.Results:1Biochemical indicators:Compared with the controlled group, in model group, the amount of serum SOD, GSH-PX activity, HDL-C have significantly different degrees (P<0.05or P<0.01); and the LPO, TG, TC, LDL-C levels also increased significantly differently (P<0.05or P<0.01). Basic the above biochemical markers, the treatment group have varying degrees improvement.2Pathological indicators:①Lungs: Compared with the control group, the alveolar volume decreases and collagen fibers and NOX2expression measured by immunohistochemistry enhanced in model group. Compared with the model group, the alveolar volume, collagen fibers, NOX2expression of treatment group improved of varying degree.②Liver:We observed extensive liver cell enlargement, fatty degeneration and collagen fibers hyperplasia in the model group. Compared with the model group, the treatment group had varying degrees lesions alleviate in liver. Compared with the control group, NOX2immunohistochemical expression would be enhanced in treatment group. Compared with the model group, NOX2expression were decreased to varying degrees in the treatment group.③rain:Compared with the control group, the cortical pyramidal and the hippocampus layer cells in the model group showed significantly increasing and NOX2immunohistochemical expression measurement enhanced. Compared with the model group, these lesions in the treatment group improved of different degrees.Conclusion:The study replicated the high-fat aging rats model through the stimulation of ozone and high-fat feeding. Three viscera injury may show that the overall characteristics the mechanism of aging model animals. YSY could reduce hyperlipidemia rat serum LPO, TG,TC,LED-C level in serum, increase SOD activity, GSH-PX,HDL-C in serum content in some degree and reduce the apoptosis of welling and nerve cells in the cerebral cortex and hippocampus vertebral layer neurons in rat hepatocytes. Also, it could reduce alveolar damage, rat liver, lung collagen fibrosis, the expression of inflammatory factors NOX2in the liver, lung, brain, and improve aging and hyperlipidemia in rat shave a therapeutic effect. The mechanism may be slowing down ageing and reducing blood fat by lowering lipid peroxide damage.
