Siwu Heji oral solution, derived from Si-Wu-Tang, a well-known composite formula of traditional Chinese medicine (TCM), consists of four herbs: Radix Paeoniae Alba, Radix Angelicae Sinensis, Rhizoma Chuanxiong and Radix Rehmanniae. Siwu Heji is widely used in clinic and also be recorded in ChP as one kind of TCM recipes. The present study provides a method to determine the index components in Siwu Heji and its consisting herbs by high performance liquid chromatography (HPLC). The integrated quality assessment of Siwu Heji oral solution and its consisting herbs was developed.(1) Several simple, rapid and reliable HPLC methods were developed to determine 5-(Hydroxymethyl) furfural (5-HMF) in Radix Rehmanniae, paeonifiorin in Radix Paeoniae Alba and ferulic acid in Radix Angelicae Sinensis and Rhizoma Chuanxiong.5-HMF in the 10 batches of Radix Rehmanniae was analyzed. The chromatography is performed on a Kromasil C18 column (250 mm×4.6 mm, 5 μm) with acetonitrile-water (95: 5) as the mobile phase and the absorbance was monitored at 284 nm. The calibration curve was linear in the range of 0.91-18.2 ×g·mL-1 with the correlation coefficient 0.9997. The mean recovery was 99.8%.Paeonifiorin in the 10 batches of Radix Paeoniae Alba was analyzed. The chromatography is performed on a Kromasil C18 column (250 mm×4.6 mm, 5 μm) with acetonitrile-methanol -water-phosphate acid (10: 20: 70: 0.035) as the mobile phase and the absorbance was monitored at 230 nm. The calibration curve was linear in the range of 0.0385-0.370 mg·mL-1 with the correlation coefficient 0.9993 .The mean recovery was 99.1%.Ferulic acid in the 10 batches of Radix Angelicae Sinensis and Rhizoma Chuanxiong was analyzed. The chromatography is performed on a Kromasil C18 column (250 mm×4.6 mm, 5 μm) with acetonitrile-methanol-water-phosphate acid (10: 20: 70: 0.035) as the mobile phase and the absorbance was monitored at 316 nm. The calibration curve was linear in the range of 0.00125-0.020 mg·mL-1 with the correlation coefficient 0.9993. The mean recovery was 97.9% (Rhizoma Chuanxiong) and 99.0% (Radix Angelicae Sinensis).
