The medullae of Juncus effusus L.was used to eliminate excess heart fi-re and to i-nduce diuresis for treatment of restlessness and insomnia etc.Its nature and flavor is sweet,bland and slightly cold, and its channel entered is heart, lung, and small intestine-e as described in Chinese medicine.The bi-ological active constituents from the medul-lae of Juncus effusus L. have b-een investigated by our research group for years.The whole Juncus effuses L.was extracted with95%ethanol at first,and then the ethanolic extract wa-s further separated with petroleum ether, chloroform, ethyl acetate and met-hanol in turn to get four fractions from large to small in polarit.Through t-he experiment of EPM and open field, we have found fraction of ethyl ace-tate have definite an-xiolytic effect.At the same time, dehydroeffusol、effus-ol、dehydrojuncusol、juncusol als-o have the effect of anxiolytic.Based on our previous study, a more effective metho-d was developed for separating phenanthrenes and the fraction of phenanthrene from the medullae of Juncu-s effuses L. Some phenanthrenes were isolated and their structure-s were el-ucidated.The anxiolytic activitie-s of the isolated phenanthrenes were evaluat-ed by Elevated Plus Maze (EPM) test.Finally, via the method of isobologr-am to study the relationship of dehydroeffusol and effusol.The dissertation included review section and experimental section.The review section was managed into three topics as following:1. the structures and biological activities of phenanthrenes from the Jun-cus effusus L2. The chemical compositions of traditional Chinese medicine arecompli-cated and diverse. When the selected one or several methods to get good e-fficicence.3. methods to study the relationship of traditional Chinese medicine.The experimental section included the isolation, structure elucidation an-d anxiolytic activity of phenanthrenes from the medullae of Juncus effusus.(1)combination silica gel column chromatography with high-speed counte-r current chr-matography three phenanthrenes were isolated. Their structures were elucidated to be dehydroeffusol, juncusol,effusol.(2)Based on our previous study, a more effective method was developed for separating the fraction of phenanthrene from the medullae of Juncus eff-uses L.(3)The HPLC fingerprints of11samples showed over90%similarity to th-ecommon pattern of HPLC fingerprint,the content of dehydroeffusol、effu sol is very high, via the method of analysis of variance to study the relati- onship of dehydroeffusol and effusol.
