Objective To observe the behavior and evaluate the levels of DA and5-HT in striatum and plasma in rat models of tic disorder(TD) induced by DPN or DOI, and provide the evidence of selecting rat models reasonably. Methods1.The rats of IDPN inducement were intraperitoneally injected IDPN, and the rats of DOI inducement were intraperitoneally injected DOI. The activity scale scores, stereotypy scale scores and classified stereotypy scale scores were observed and recorded by double-blind method.2. To observe the levels of DA and5-HT in striatumin and plasma of the rat models of TD induced by IDPN or DOI by ELISA, and discuss the mechanism of the rat models.Results1.The activity scale scores, stereotypy scale scores in the IDPN (the activity scale scores:1.20±1.03,1.50±0.97,2.20±1.14,3.20±1.40; stereotypy scale scores:2.10±1.20,2.20±0.63,2.10±0.88,2.40±0.52) and DOI (the activity scale scores:1.40±0.89,1.80±0.45,1.80±1.10,2.20±0.45; stereotypy scale scores:2.00±1.23,1.60±1.14,2.40±0.55,2.00±0.71) inducement groups were higher than that in the control group(the activity scale scores:0.20±0.43,0.30±0.68,0.40±0.70,0.70±0.95; stereotypy scale scores:0.30±0.68,0.40±0.70,0.40±0.84,0.60±0.84) at1d-4d(F=5.32~17.52, P<0.05). And all classified stereotypy scale scores except grooming and cage licking in the IDPN(2.60±1.65,2.16±2.70,0.47±0.77,2.50±1.84,12.86±7.06,1.29±1.78) and DOI(6.71±3.33,0.66±0.99,0.54±0.82,0.11±0.45,4.40±2.66,0.28±0.57) inducement groups were higher than that in the control group(1.39±1.24,0.72±1.02,0.06±0.24,0.05±0.01,0.02±0.02,0.15±0.11)(F=2.85~50.57, P<0.05).The activity scale scores of rat in the IDPN inducement group(3.60±1.27,3.80±0.63,3.60±0.84,2.80±0.45,2.60±0.55,2.40±0.55) were higher than those in the control group(0.40±0.52,0.60±0.84,0.50±0.71,0.40±0.74,0.80±0.44,0.40±0.53) and DOI group(1.20±0.84,1.60±0.55,2.00±0.71,1.40±0.30,0.80±0.84,0.60±0.90) at5-7d,14d,21d and28d; the stereotypy scale scores of rat in the IDPN inducement group(2.30±0.48,2.30±0.95,3.60±0.73,2.80±0.14,3.20±1.54) were higher than those in the control group(0.50±0.71,0.50±0.85,0.80±0.63,0.40±0.55,0.60±0.66) and DOI group(1.20±1.10,0.80±0.84,0.60±0.90,0.80±0.92,0.80±1.10) at5d,7d,14d,21d and28d(F=11.15~50.70, P<0.05).The classified stereotypy scale scores of wood chip eating,circling,head shaking and choreoathetoid movement of rat in the IDPN inducement group(2.16±2.70,2.50±1.84,12.86±7.06,1.29±1.78) were higher than those in the DOI group(0.66±0.99,0.11±0.45,4.40±2.66,0.28±0.57); the classified stereotypy scale scores of repetitive paw-to-mouth movements and self-gnawing of rat in the DOI inducement group(6.71±3.33,0.54±0.82) were higher than those in the IDPN group(2.60±1.65,0.47±0.77),(F=2.85~50.57, P<0.05).2. The DA concentration in plasma was higher in the IDPN(7.70±3.12) and DOI groups(1.01±0.74) than in the control group(0.32±0.12)(F=13.43,P<0.05).The DA concentration in striatum was lower in the IDPN model group (68.13±12.34) than in the DOI group(98.56±21.56) and the control group (137.45±20.14)(F=8.60, P<0.05). The5-HT concentration in striatum was found to be lower in the DOI model group (56.83±34.72) compared to the IDPN model group (78.52±10.03) and control group (109.14±14.05)(F=3.65, P<0.05).Conclusions1. The generalized tics including circling, head shaking and choreoathetoid movement were obviousely observed in the IDPN models. The partial tics including repetitive paw-to-mouth movements and self-gnawing were obviousely observed in the DOI models. The IDPN models and the DOI models can perfectly copy the high excitement, high activity and tic behavior of TD patients.2. The decreasing DA concentration in striatum of the IDPN models could lead to increase of DA uptake from the synaptic cleft. The exciting directly5-HT receptor of the DOI models could effect DA system and lead to tic disorders.3. The IDPN models are shorter-time cost, longer persistence and lower cost than the DOI models.
