Objective:To inhibit the expression of ATF5gene in nude mice by ATF5siRNA carrie d lentiviral vector,and to observe its effect on the growth, apoptosis of ovarian c ancer transplantation tumor in nude mice.Methods:1.Ovarian cancer cell line SKOV3was injected into nude mice to structure xe nograft tumor model.Seven days after inoculation,30nude mice were segmented int o3groups randomly.distinct treatment was given for different groups:blank cont rol group:PBS was injected into tumor;negative control group:negative lentivirus ve ctors was injected into tumor;experimental group:lentivirus-delivered ATF5siRNA was injected into tumor.The growth of tumor was observed at the same time.The volume of these tumors was measured,then the curve of tumor growth was describ ed.After four weeks,the nude mice was killed.The weight of tumor was measur ed,and the inhibition rate was calculated.2. The expression of ATF5was detected by western-blot and RT-PCR.The a poptosis of tumor tissue was examined by TUNEL.Results:1. The growth rate of transplantation tumor significantly ruduced in experimen tal group,and the tumor’s weight and volume in experimental group were lesser th an those in blank control group and negative control group significantly.2. The expression of ATF5in experimental group were significantly decreased, which showed the expression of ATF5gene can be significantly silenced by lent1v1rus. 3. TUNEL showed the apoptosis of transplantation tumor increaased significan tly after silencing ATF5gene.AI of experimental group was34.63±1.91%,negative control group6.04±0.19%,blank control group5.07±0.20%.There is obvious diff erence between controls and experimentalConclusion:The lentivirus-delivered ATF5-siRNA can markedly inhibit the expression of ATF5, effectively inhibit the growth of xenograft tumor,and promote the apoptos is of xenograft.ATF5is expected to be a potent target of gene therapy in ovarian cancer.
