In this paper,Camellia oleifera seed shell and Camellia oleifera fruit hull(the polysaccharides from seed shell are called CSP and that from fruit hull are called CFP)were taken as raw material,constituents of which were determinated.In addition,tea saponin from CSP and CFP were succesively extracted,seperated and purified.Compostion of them were also studied.The main contents of this paper were the following parts:(1)The principle components of CSP and CFP,such as tea saponins, had fat, total nitrogen, total sugars, reducing sugar, total flavonoids and cellulose were determined with chemical methods.The contents of CFP is 10.27%、0.91%、2.84%、10.75%、2.46%、15.67%、2.59%,and the contents of CSP is19.26%、1.93%、1.53%、8.43%、2.11%、14.85%、3.92%。(2)Tea sapionins were extracted from CSP and CFP though twe methods, water extraction and alcohol extraction.The optimum condition of water extraction process from CSP was that the solid-liquid ratio of 1:12,extraction temperature of 75℃,extracting time of 3h;The optimum condition of water extraction process from CFP was that the solid-liquid ratio of 1:14,extraction temperature of 80℃,extracting time of 3h;The optimum condition of alcohol extraction process from CSP was that the concentration of ethanol of 95%,the solid-liquid ratio of 1:12,extraction temperature of 70℃,extracting time of 3h;The optimum condition of alcohol extraction process from CFP was that the the concentration of ethanol of 95%,solid-liquid ratio of 1:12,extraction temperature of 70℃,extracting time of 3h;The tea sapionins by alcohol extraction had ligter color and higher purity,and with little bubble during concentration,so use this kind of tea sapionins to purify.(3)Effect of different decolorizers on tea saponin decolorization was studied.the optimum condition of hydrolysis found by orthogonal test was:decoloring temperture 60℃,decoloring time 90 min,3%H2O2,PH=10. With the material of pretreated alcohol extract of CFP and CSP,the adsorption and desorption effects on tea saponin of macro porous resins were studied.The results showed that D101 was the suitable resin of purifying tea saponins.The results showed that the optimum adsorption conditions were the feeding rate of 3BV/h,sample concentration of 20mg/m L, Under these conditions: the tea saponin adsorption rate was 68%.The optimum desorption parameters at the eluent alcohol concentration of 85%, the flow rate of 2.0 BV/h,the eluent volume 5.0 BV.As a result, tea saponin desorption rate was 82%.All the datas shows that the optimization was feasible. The purified tea saponin had significantly improved the purity. The experimental results provided a theoretical basis for the future industrial production of purified tea saponin.(4)The CFP,CSP and Camellia cake tea saponins were treated with a series of test such as color reactions,foamability,.The three kind of tea saponins were characterised by two features color reaction. One was that the reaction of tea saponins and 10% α-naphthol-ethanol solution present a purple ring. The other was that when tea saponins reacted with acetic anhydride, at the beginning the mixed solutions was separated and then displayed red in the shaking, at last turned purple after standing for a long time.Its high foaming ability and foam stability made tea saponins be applied in washing chemical industry and building materials industry field.Their structure was analyzed by UPLC/Q-TOF MS, the saponins were forecasted to be theasapomins A2, camelliasaponins B1 and camelliasaponins B2, assamsaponins F.
