Studies have shown that collagen extract from tilapia fish scale is mainly Type-Ⅰ collagen,but which from pearl oyster mantle is type-Ⅴ like collagen,they are different in composition,arrangement and structure on amino acid.Currently,studies on the extraction and use of scale and the mantle of collagen has been widely reported,but the two types of collagen comparative study has not yet been reported.In this paper,by comparing the two types of collagen,we determine which type is more suitable for the preparation of antioxidant peptides and collagen peptide chelated calcium products, witch provide a theoretical basis for the development and utilization of pearl oyster mantle collage,and studding the influences of calcium chelating and antioxidant capacity of collagen.The main contents are as follows:1Extract of Tilapia fish scales and the mantle of pearl oyster collagen and amino acid analysisUsing hot water extraction,tilapia fish scales and pearl oyster mantle collagen was extracted,when tilapia fish scale extraction of collagen,heat soluble collagen was yield of3.5%,thermal insoluble collagen was6.8%;pearl oyster mantle extract collagen,heat soluble collagen was yield of5.6%,thermal insoluble collagen was3.2%.After a comparative analysis of amino acids,found that amino acid composition of the heat extraction collagen from two materials mentioned above are consistent with the basic features of the amino acid composition of collagen,but compared to some amino acids have a significant difference between the two materials.In scale collagen,alanine,lysine and proline content than the mantle collagen,and collagen in the mantle,glutamic acid, valine,methionine and isoleucine higher than scale collagen.2Anti-oxidant activity comparison of tilapia fish scales and pearl oyster mantle collagen peptideRespectively,alkaline protease,neutral protease,acid protease and papain enzyme for both raw materials obtained peptide,these four enzymes,either enzyme digestion,pearl oyster mantle collagen peptide DPPH scavenging and hydroxyl radical scavenging rate was higher than the same kind of scale obtained by enzymatic hydrolysis of collagen peptide,and after digestion to reduce the molecular weight antioxidant activity of collagen was significantly increased,indicating that the molecular weight polypeptide associated with antioxidant activity.Mantle collagen enzymatic with alkaline protease has the highest DPPH clearance rate and hydroxyl radical scavenging witch reached34.4%and71.2,respectively. Tilapia fish scale mantle with SEPHADEX G-25and the separating alkaline protease papain-treated appeared peaks respectively four and two components,six components collected from the peak,respectively MY-1,MY-2and JW-1,JW-2,JW-3,JW-4.Analysis found that the antioxidant component MY-1activity is higher than MY-2,JW-1is the highest antioxidant activity component in JW-1,JW-2, JW-3,JW-4four components.Respectively with pepsin,chymotrypsin and trypsin highest antioxidant activity components JW-1in vitro simulated digestion experiments and found that after in vitro digestion test,the antioxidant activity was almost unchanged.3Comparison of calcium chelating ability about tilapia fish scales and pearl oyster mantle collagen peptide.Four enzymes were digested with scales and mantle collagen,found that in either enzyme digestion,calcium chelation rate of pearl oyster mantle collagen peptides have been higher than the same kind of scale enzymatic hydrolysis of collagen peptide.And in the four enzymes,papain liquid calcium chelation rate is higher,which is28.2%scale,the mantle of34.5%.Separated by ultrafiltration system,will be in the same type of collagen hydrolyses calcium chelating ability of collagen peptides were isolated5000Da above(two)and5000Da the following(two)four components,compared calcium chelating ability of these four components found in the highest molecular weight5000Da less papain mantle collagen chelate rate,witch reached39.8%.4Calcium chelating optimization of component which molecular weight lawer than5000Da of pearl oyster mantle collagen peptide extracted by papain enzyme digestionUsing single factor test to determine the factors affecting collagen peptide chelating calcium rate,then the orthogonal experiment method,the molecular weight5000Da following papain pearl oyster mantle collagen peptide optimized chelation process.The optimum conditions for the calcium chelation collagen peptides as follows:pH is8,the collagen peptide with a mass ratio of4:l;temperature is50℃,chela.ting20min,under the conditions calcium chelating ratio is highest which reached40.2%.
