Chirality is one of the fundamental properties of nature,the activity of chiralcompounds are often different.Chromatography is the most main chiral analysistechnology,but in the analysis test of chiral samples, application of chromatographytechnique chiral analysis is limited by the lack of optical detector.Optical detectiontechnology can distinguish between the symmetry chiral compounds, Combined withchromatographic detection technology can realize the separation of chiralcompounds,so it plays an important role in the field of biological pharmacycomposition analysis and so on. This paper is from the principle and technology ofoptical detection system research and analysis, and aims to demarcate and test theoptical detection system.The theoretical and experimental research shows that the first question in theproduction of optical detector is the choice of light source, because differentwavelength optical rotation rate is different,so the choice of light source for opticaldetector is very important. Experiment mainly combined laser, LED light source,incandescent lamp, incandescent lamp with filter to compare, finally decided tochoose small volume, simple drive power supply, cheap price, goodmonochromaticity of LED light source as the optical detector’s light source. Heliumneon laser’s volume is too big,it isn’t conform to the small volume requirement ofthe experimental source. The human eye is more sensitive to green, so choose thewavelength of555nm green LED light source.Instrument’s hardware based on common optical rotation detector system,electrical system, the structure of the machine frame and software design,through thedesign of deflection angle counter module, finally combined into deflection angleand the time relationship of online separation of chiral compounds opticaldetector.Instrument’s software programming based on Delphi7,pure water and twokinds of glucose solution are used to detect the optical phenomenon, using curvefitting’s least square method processing data and determine the polarization angle ofthe three samples,quadratic fitting and one fitting two kinds of algorithms are analyzed in detail.Experiments have shown that one fitting is simple than quadraticfitting, two kinds of algorithms’ accuracy is very close,but overall considerationfrom the experiment measurement speed and accuracy, a fitting is more suitable thanquadratic fitting for the optical detection experiment.In order to verify the accuracy of the system test, Experiment using self-makechiral detector and WZX-1optical degrees overhead light meter measure L-tyrosinesolution respectively, Calculate the mean value of the rotation angle of L-tyrosinesolution, and comparing with standard value,verified the accuracy of systemdetection.Respectively collect pure water, low concentration,high concentration ofglucose solution and L-tyrosine solution for20groups data,exmine the systemmeasurement’s stability and accuracy to chirality material.Then respectivelycontinue detect pure water, low concentration,high concentration of glucose solutionand L-tyrosine solution, verified the system online continues measurement’s stabilityand accuracy to chirality material.It is proved that the system of optical material detection accuracy can meet theneeds of the experiment, and can be directly clear read the material of the rotationAngle in the user interface, which can accurately identify the substance isleft-handed or right-handed.
