In order to obtain high-quality Agaricus blazei Murrill polysaccharides (ABMP),this study investigated the effects of extraction and drying methods on polysaccharideyield, chemical composition and antioxidant activities of ABMP. After that, the masstransfer kinetic model of polysaccharides extracted by hot water was established.The experimental condition of different extraction methods was conducted basedon single-factor experiments. Five polysaccharides were extracted from Agaricusblazei Murrill (ABM) through different methods. Their chemical and physicalcharacteristics were determined and antioxidant activities were investigated on thebasis of DPPH radical assay, hydroxyl radical assay, ABTS radical assay,Fe2+-chelating and reducing power. The results showed that the ABMP extracted bycompound enzymes was found to present the highest polysaccharides yield (17.44%),and ABMP extracted by hot water showed the lowest polysaccharides yield of11.95%,the content of uronic acid was31.23%and27.43%. The content of uronicacid showed the similar trend of polysaccharides yield. Compared with other methods,ABMP extracted by compound enzymes exhibited the strongest antioxidant activity,On the contrary, hot water extracts exhibited the lowest antioxidant activities. Onhydroxyl radical scavenging assay, the IC50value of polysaccharides extracted bycompound enzymes and hot water was398.92μg mL-1and810.68μg mL-1. ThreeABMP extracted by single enzyme exhibited no significant difference inpolysaccharides yield and antioxidant activities.According to above experiments, the effects of drying methods onphysiochemical properties and antioxidant activities was carried out. Results showedthat the three ABMPs have different physicochemical and antioxidant properties.Compared with air drying and vacuum drying methods, freeze drying method resultedin ABMP with the higher content of neutral sugar, uronic acid and higherpolysaccharides yield and antioxidant abilities.Finally, according to Fick’s second law, the mass transfer dynamic model of hotwater extraction method was established, the data fitting the equation.
