The natural bioactive flavonoids widely exist in nature which has obvious curative effect for human diseases.In the use of flavonoids,people have been pursuing to improve the curative effect and reduce its side effect.The most ideal method should be to influence drug targeting to the diseases of organs,tissues or cells. Differences of flavonoids structure influence their transportion,absorption,metabolism and biological activity in human body.Regardless of flavonoids as small molecules directly or indirectly into the blood stream can combine with plasma proteins and the bonding degree is closely related to the structure of polyphenol.With cross integration of nanometer technology and biomedical research,how to combine nanotechnology to enhance biomedical research methods and means and to achieve new breakthrough research level have become the important objectives of the development of nanobiomedical.In this work,using of Bovine serum albumin as a carrier protein and 32 different structure of flavonoids formed 32 BSA/ flavonoids nanoparticles. Structure of polyphenols has an effect on encapsulating of BSA and flavonoids and the antioxidant activity change of being encapsulated flavonoids.The main research contents are as follows:Firstly,characterization of three kinds of nanoparticls morphology and BSA were photoed by TEM, the result showed that three kinds of nanoparticls were 20 nm and the structure of BSA had not be changed.Secondly,32 kinds of BSA /flavonoids nanoparticls encapsulatin rates were analyed by HPLC,the results showed that the encapsulatin rates of 32 nanoparticls were obvious different. The structure of flavonoids was very important for the effect of BSA and flavonoids.Combination of small molecules and protein interaction relationship analysis conculuded that flavonoids coating glucoside hydroxyl had a weak interaction with BSA results in low encapsulatin rate. Flavonoids coating hydroxyl methylation and B ring number of hydroxyl groups had a strong interaction with BSA results in high encapsulatin rate. Hydroxyl position of flavanone affected its interaction with BSA and encapsulatin rate was different. Thirdly,strong antioxidant activity of 12 kinds of flavonoids were selected in thisparper, the antioxidant activity of encapsulated flavonoids comprehensively was evaluated by three kinds of antioxidant experiment system.The results showed that the antioxidant activity of encapsulated flavonoids decreased comparing with free flavonoids in the same concentration.By IC50 reflected that different flavonoids in the three antioxidant experiment of weaking degree was also different. The 11 kinds of flavonoids forming part of the location of hydrogen bonds would be estimated by combining with the characteristics of three kinds of antioxidant experiment preliminarily.
