This paper mainly studies an online purification and quantitative method for water soluble vitamins in complex matrix. The target is VB1, VB3, VB6, VB12, VB2, VB5, VB7 and Vc.The first chapter of the paper expounds on the importance of vitamin determination of the daily food and vitamin supplements, as well as the commonly used determination method and sample pretreatment method. The second chapter mainly introduces the high performance liquid chromatography and two-dimensional chromatography heart-cutting technology. The third chapter elaborates the two-dimensional chromatography heart-cutting system building. A two-dimensional liquid chromatography system with heart cutting technology, based on the Thermo Dionex Ultimate double ternary liquid chromatographic system, develop a one-dimensional purification in removing impurity, two-dimensional quantitative the target vitamin method.8 target vitamins should be cut from the first dimension to the next dimension, and a precise and smoothing running valve switching method is needed. The thesis examines the combination model of a variety of column, the final choose, one-dimensional is Acclaim PA, two-dimensional use Acclaim C18 column to separate and analysis 8 target vitamins, got good linear, reproducibility and recovery. In real sample analysis, VB1, VB5, VB6 and VB2 are detected, ultraviolet spectra corresponding is good, and meet the marked quantity. VB3 and VB7 were not detected, but the standard addition recovery rate is good. Vc content in the sample is very high, can be detected, but not quantitative in this paper because of its instability. VB12 content is very low and there is co-elution interference, failed to check out. But under another column combination, one-dimensional Hypersil Gold Phenyl, two-dimensional Acclaim C18, VB12 can be baseline separated, and accurate quantitative.In this paper, we provide an automatic, efficient, repeatable pretreatment method, and an accurate, repeatable and good recovery determination method for the water soluble vitamin in complex matrix sample. And this two-dimensional heart-cutting method also can be applied to the determination of other complex matrix.
