This study established a UV–Vis spectrometry method to determine the lignans content in flaxseed oil which were extracted by flaxseed as raw material, studied the impact of unit solvent and multi-solvent to flaxseed oil extraction rate and the flaxseed oil content of lignans, the processes were also optimized, and the suitable process conditions of solvent extraction of flaxseed oil rich lignans.According to the structural properties of lignans, as well as the differences between the polarity of faxseed oil, UV–Vis spectrometry were adopted to determine the content of secoisolariciresinol diglucoside(SDG) of flaxseed oil. SDG had the characteristics of absorption at 200~400 nm ultraviolet region, the spectral scanning curves showed the max absorption of wavelength was 280 nm, were got by drawing standard curve the significant linear relationship between concentration of SDG and absorbance. According to the principle of “similar dissolve mutually”, oscillating extracted SDG of linseed oil with methanol, determined absorption, by the regression equation calculate content of SDG and rule out other possible interference substances. The results showed that: the method was simple, fast, stable, average recovery was 99.7%, RSD was 1.9%. According to the structure properties and content of flaxseed lignans, this method could be used to determine total lignan content of flaxseed oil.According to the polarity of organic solvent, acetone, ethyl acetate, isopropanol, isopropanol / water, ethyl acetate / ethanol, hexane / ethanol were elected as solvents of extract lignan-rich flaxseed oil, extraction temperature, extraction time, ultrasonic power, the ratio of material to solvent as factors, with flaxseed oil extraction rate and lignan content of flaxseed oil as test indicators to screen solvents, It was found that the extraction solvent acetone was ideal lignan-rich flaxseed oil.The extraction of flaxseed oil with acetone was optimized by Design-Expert 8.0.6.0. The mathematical models of the key factors affecting flaxseed oil extraction rate and flaxseed oil content of lignans were established. The result showed that the optimum conditions were extraction time 38 min, extraction temperature 48℃, ultrasonic power 210 W, the ratio of material to solvent(g/m L) 1:9. Under this conditions, flaxseed oil extraction rate was 94.51%, lignan content of flaxseed oil was 0.69%.
