The Krill oil extracted from Antarctica krill is rich in phospholipid,astaxanthin and omega-3 polyunsaturated fatty acids(w-3 PUFA) such as eicosapentaenoic(EPA, 20:5n3) and docosahexaenoic(DHA, 22:6n3) acids. Krill oil have various potential health benefits including promotion brain development, protection against diabetes and cardiovascular disease(CVD), anti-aging, and so on. Krill oil was extracted by organic solvent extraction, Supercritical carbon dioxide extraction and enzymatic extraction. Objective:1)To optimize organic solvent extraction progress for krill oil.2)To build and optimize enzymatic extraction for krill oil.3)To research the method of enrichment of phospholipid from Krill oil. Method:1)Refining the oil extracted by ethanol extraction to reduce insoluble impurities content of the oil.2)Researching the effect of the conditions in hydrolysis and extraction process for extraction efficiency by single factor experiment and response surface analysis.3)Enrichment of phospholipid by Fractionation technology. Result:1) The receiving rate of Krill oil extracted by 95% ethanol were 21.26% before refined and 15.28% after refined, the phospholipid content were 28.22% and 41.02%, astaxanthin 177 ppm and 250 ppm, moisture and volatile 4.46% and 1.51%, insoluble impurities 27.80% and 0%; The receiving rate of Krill oil extracted by absolute ethanol were 20.51% before refined and 16.33% after refined, the phospholipid content were 31.69% and 42.19%, astaxanthin 197 ppm and 375 ppm, moisture and volatile 4.39% and 1.49%, insoluble impurities 20.09% and 0%;2) The extraction efficiency of oil extracted by enzymatic extraction was highest under the conditions: Enzyme dosage 0.13%, temperature 43.8 ℃, time 4.9 hours in hydrolysis process; ethanol dosage 0.74 times, Petroleum ether 0.9 times, time 43.22 min in extraction process; velocity 2000 rpm, time 10 min.3) The phospholipid content was increased respectively to 39.53%, 41.19%, 44.69% and 51.02% when krill oil extracted by enzymatic extraction using boiled krill as raw material were respectively deal with Fractionation technology in 8 ℃, 4 ℃, 0 ℃,-20 ℃.Conclusion: 1) The refining method was effective based on the result that the insoluble impurities of krill oil refined were wiped off.2) The optimal conditions of enzymatic extraction technology were : Enzyme dosage 0.13%, temperature 43.8 ℃, time 4.9 hours in hydrolysis process; ethanol dosage 0.74 times, Petroleum ether 0.9 times, time 43.22 min in extraction process; velocity 2000 rpm, time 10 min.3) The optimal conditions of Fractionation technology were : temperature 0 ℃, time 9 h.
