Magnetic targeting drug carrier can be gathered with the blood circulation to the target tissue and released targeted drugs to treatment of cancer but does not damage the body’s normal cells. Liposomes have been widely used as biomedical materials because of their good biocompatibility. In this thesis, AuNPs, FeAunano-alloys were successfully prepared by a simple sodium bis(2-ethylhexyl) sulfosuccinate (AOT)-octane-water(W/O) microemulsion system. AuNPs, FeAunano-alloys were encapsulated in liposomes, and its properties were characterized. The results are as follows:1. Preparation and Characterization of AuNPsAuNPs with different sizes were prepared in the n-heptane/AOT/water inverse microemulsions, and were fully charactered by XRD, EDS, TEM, DLS, VSM,Uv-Vis. The results showed that AuNPs had good spherical shape and narrow size distribution, there was a typical absorption peak at519nm in the UV spectra.It could be due to the size of AuNPs can be controlled by the molar ratio of water to surfactant AOT in the microemulsion.2. Preparation and Characterization of FeAu nano-alloysFeAu nano-alloys modified by dodecanethiol with different sizes were prepared in the n-heptane/AOT/water inverse microemulsions, and were fully charactered by X-ray powder diffraction (XRD), X-ray energy dispersive spectroscopy (EDS), transmission electron microscopy (TEM), dynamic light scattering instrument (DLS), UV-visible spectrophotometry photometric properties (Uv-Vis), Superconducting quantum interference device (SQUID), Fluorescence spectrophotometer were characterized. The results showed that FeAu nano-alloys modified by dodecanethiol has good spherical shape, single dispersivity and narrow size distribution, it also have magnetic response at room temperature. In the UV-Vis absorption spectrum, the absorption peak of S-band of FeAu nano-alloys has a obvious red shift in comparison with traditional AuNPs. The UV-Vis absorption peak can red shift to615nm whlie the size is12nm. Therefore, controlling in the form of water core radius of the microemulsion system must be exerted to control the size of the FeAu nano-alloys.3. Preparation and Characterization of Berberine hydrochloride magnetic targeting-photosensitive liposomesMagnetic targeting-photosensitive liposomes were prepared by super critical CO2 method and the size can be controlled by CO2pressure. The morphology and size of magnetic targeting-photosensitive liposomes were fully charactered by TEM. The results showed that the sizes of magnetic targeting-photosensitive liposomes were80-200nm. Encapsulation ratio, encapsulation yield and in vitro release of drug were measured by UV method. Results indicated that encapsulation ratio and encapsulation yield decreased with different contents of Fe. The effects of Fe at different contents, the time of UV irradiation on magnetic targeting-photosensitive liposomes were studied. The released efficiency is attained about75%and66%.In this study, we have done a useful exploration orienting effect, on the photocontrolled release behavior of liposomes. It can achieve a greater breakthrough in creativity, and it has tentatively probed in this aspect.
