Dietary fiber is the seventh largest human nutrients, it has lower serum cholesterol, promote gastrointestinal health, prevention of high blood pressure, colon cancer, obesity, heart disease, atherosclerosis and other physiological functions. In this paper, dietary fiber extraction conditions were optimized using the amino acid method and three-sodium dietary fiber Amine sulfated modification research hypoglycemic activity of dietary fiber before and after sulfation. The main contents and results are as follows:(1)Black bean dregs insoluble dietary fiber extraction conditions optimized. Single factor test to determine the extraction conditions for solid-liquid ratio 1:20, NaOH concentration of 0.15mol/L, ultrasonic temperature 60℃, ultrasonic power 150W, ultrasonic time 30 min, on the basis of single factor experiments, the use of orthogonal test reaction condition. OK insoluble dietary fiber extracted from the optimal reaction conditions:solid-liquid ratio 1:20, NaOH concentration of 0.2 mol/L, ultrasonic temperature 60℃, ultrasonic power 150 W, ultrasonic time 40 min, under optimal conditions of protein removal 95.09%.(2)Preparation of black bean dregs amino acid insoluble dietary fiber sulphate. To the degree of substitution (DS) as an indicator to study amino acid amount, ultrasonic temperature, ultrasonic power, ultrasonic time on the DS. Based on single factor experiments, response surface optimization of reaction conditions, the test to determine the black bean dregs insoluble dietary fiber sulfate optimum conditions for:amino acid addition of 1.96 g, ultrasonic temperature 78.84℃, ultrasonic power 249.93 W, ultrasound time 34.84 min, to obtain a DS of 0.681 under this condition.(3)Three sodium black beans prepared amine legal dregs insoluble dietary fiber sulphate. To the degree of substitution (DS) of study factors, selecting NaHSO3 and NaNO2 molar ratio of esterification agent preparation temperature, temperature esterification agent time to prepare single-factor test, on the basis of single factor experiments, response surface optimization of reaction conditions for test to determine the optimum conditions for:nNaHSO3/nNaNO2= 4.29, prepared by esterification agent temperature 91℃, esterification agent preparation time 91 min, get DS to 1.193 under this condition. To the degree of substitution (DS) of study factors, choose the amount of water, the sample amount, pH, ultrasonic temperature, ultrasonic time, ultrasonic power single-factor test, on the basis of single factor experiments, the use of orthogonal experiment to optimize the reaction conditions, to determine the optimum conditions for preparation:Add water 15 mL, sample amount 5 g, pH value of 9, ultrasonic temperature was(4)45℃, ultrasonic time of 40min, ultrasonic power is 250 W. To obtain a DS of 1.273 under this condition.(5)Black bean dregs insoluble dietary fiber sulphate hypoglycemic research. The results showed that black bean dregs insoluble dietary fiber AGC presence of sulfate inhibition, and increase their concentration and AGC inhibition with enhanced degree of substitution. Black bean dregs insoluble dietary fiber sulfate inhibition of AGC is reversible inhibition by double reciprocal curve drawn as competitive inhibition.
