Banana postpartum processing level is low,the industrial chain is short.In order to promote the development of post-production deep processing of bananas,it is necessary to develop banana related active ingredients.At present,the research is mainly limited to the optimization of the extraction process of dietary fiber from banana skin and the runchang purge function of banana pulp.However,There is little research on the extraction technology of banana pulp dietary fiber(BP-DF).The mechanism of regulating gut microbiota(GM)is not clearIn this paper,using banana pulp as raw material,the optimal extraction process was optimized through single factor test and response surface test,and the high-purity BP-DF was obtained,at the same time its structure and functional characteristics were studied.The effects of BP-DF on GM and intestinal function in normal mice and the regulatory effects of BP-DF on metabolic disorders and GM disorders in high fat diet(HFD)mice were investigated.The main conclusions are as follows:(1)Optimize the condition of removing starch and protein from raw materialsThe optimal conditions for the removal of starch were:compound enzyme amount8.2%,reaction temperature 60℃and reaction p H 6.The optimal conditions for removing proteins were:protease amount 3%,reaction time 93 min,reaction temperature 40℃,reaction p H 7.Furthermore,it was determined that the order of eliminating starch and then protein was better.Finally,the starch and protein elimination rates were 95.88%and 94,respectively.Banana pulp soluble dietary fiber(BP-SDF)and Banana pulp insoluble dietary fiber(BP-IDF)content were 7.61%and 16.84%respectively.(2)The physicochemical properties and antioxidant properties of banana pulp dietary fiber components were analyzedPectin in BP-SDF accounted for 57.03%,cellulose,hemicellulose and lignin in BP-IDF accounted for 37.88%,21.65%and 17.56%,respectively.The content of 6monosaccharides in BP-SDF was significantly higher than that in BP-IDF.The structure of BP-IDF is loose and porous,and presents a honeycomb network.The surface of BP-SDF is rough and irregular,and presents a flake.The crystallinity and thermal stability of BP-IDF are higher than that of BP-SDF.The FT-IR of both showed characteristic absorption peaks of polysaccharides.In addition,both BP-IDF and BP-SDF show good hydration performance.The oil holding capacity of BP-IDF is stronger than that of BP-SDF.Functional characteristics showed that BP-SDF adsorbed glucose,NO2-,cholesterol and antioxidant capacity better than BP-IDF.(3)Effects of banana pulp dietary fiber on gut microbiota in normal miceCompared with normal mice,after BP-DF intervention,the body weight and food intake of the mice decreased,and the fecal wet weight increased.Levels of p H and ammonia decrease and water content increases in faeces.According to the colon biopsy,the muscle thickness and mucosal thickness of the mice increased after bp-df intervention.In addition,BP-DF can increase the diversity and richness of GM in mice,promote the growth of probiotics,and inhibit the growth of harmful bacteria.Different BP-DF components affect different kinds of GM.Analysis of Kyoto Encyclopedia of Genes and Genomes(KEGG)showed that the BP-DF component could reduce the pathogenicity of microorganisms in the intestinal tract.(4)Effects of banana pulp dietary fiber on metabolic disorders and gut microbiota disorders in high fat diet(HFD)miceThe metabolic indexes of HFD mice were out of balance:body weight increased,the level of blood lipid and liver fat increased,severe fatty liver phenomenon,and the content of short chain fatty acid(SCFAs)in feces significantly decreased.After BP-DF intervention,all metabolic indexes were significantly relieved.The abundance of Firmicutes increased,the abundance of Bacteroidetes decreased,the diversity and richness of GM decreased,the growth of probiotics was inhibited,and the increase of pathogenic bacteria caused GM disorder in HFD mice.After BP-DF intervention,the change of GM caused by HFD was reversed.KEGG function prediction found that the pathogenicity of HFD mice increased,and the pathogenicity decreased after BP-DF intervention.
