Through domestication and screening Rhodopseudomonas palustris G3, which is selected and saved by laboratory, the writer got highly- resistant to landfill leachate Rhodopseudonas palustris G3-01, and further study the culture conditions and methods about this bacteria, preliminary discuss the possibility of using the landfill leachate to culture Rhodopseudomonas palustris G3-01, and study it’s effect on Wuxi Huilian garbage Thermal Power Plant’s landfill leachate.First put R. palustris G3 to increasing concentrations of landfill leachate for domestication then select the R. palustris G3-1 from highly resistant landfill leachate via dilution plate coating. Optimize the culture conditions such as PH, loaded volume, inoculum size and temperature, and the best culture conditions were optimized by single factor test, it is pH 8.0, loaded liquid amount 100mL/250mL, temperature 30℃, and inoculum size 6%.According to the characteristics of R. palustris could grow in light of anaerobic and darkness aerobic conditions, and preliminary study the R. palustris G3-01 two-step fermentation, we can get the best optimal fermentation culture program through the test: dark aerobic incubation 2d, light anaerobic culture 5d; based on R. palustris G3-01 flask test, the writer go on 25L fermenter small pilot test, R. palustris G3-01 growth peaked was founded in 4d. The bacteria number of R. palustris G3-01 was tested out to be 16000000000/mL, after 7d culture.Try to use the landfill leachate to culture R. palustris G3-01, we can get better cost-saver fermentation medium of landfill leachate (landfill leachate diluted five times) (g/L): anhydrous sodium acetate 0.2, yeast extract 0.3, ammonium sulfate 0.2, sodium lactate 0.2.R. palustris G3-01 landfill leachate was used for processing Wuxi Huilian garbage Thermal Power Plant landfill leachate. The test proved under dark aerobic conditions, Rhodopseudomonas palustris G3-01 had good effect on landfill leachate COD, ammonia nitrogen and total phosphorus. The highest COD removal rate reached 70.62%, the highest ammonia nitrogen removal rate reached 55.56%, the highest total phosphorus removal rate reached 68.84 percent, the test also confirmed that R. palustris G3-01 which was cultured by landfill leachate fermentation can be used in the treatment of landfill leachate.
