Fluorescent molecular probe, because of its specific selectivity, high sensitivity, simple operation, less time consuming, good bioconpatibility and other advantages, is widely used in biological sciences, preclinical medicine, environmental monitoring and other fields. Reactive oxygen species play important roles in a diverse range of physiological and pathological processes, and have close relation with many enzymes in living organisms. In this paper, we designed and synthesized three fluorescent molecular probes for the detection of hypochlorite, glucose and acetylcholine/ pesticides based on the detection of reactive oxygen species, and its application were studied. The main contents are summarized as follows:(1) A simple fluorescent probe 1 bearing 7 – nitrobenz-2 – oxa-1,3 – diazole and selenomorpholine was designed and synthesized. The probe showed highly selective, sensitive and fast(< 10 s) recognition to hypochlorite in aqueous solutions, with a low detection limit of 3.3 nM. Futhermore, the probe was capable of monitoring hypochlorite reversibly in the presence of glutathione – hypochlorite, and has been used for determination of hypochlorite in living cells with satisfactory results.(2) A new naphthalimide derivative 2 containing hexanoic acid and boronate groups that could colorimetric and fluorescence detect hydrogen peroxide was designed and synthesized. Based on the indirect detection of hydrogen peroxide enzymatic glucose, the probe could achieve the purpose of detecting glucose in 100% water. Lab studies showed the probe had the advantage of excellent selectivity, high sensitivity and low detection limit(0.3 μM). Moreover, the fluorescent probe has been used for determination of glucose in serum samples with satisfactory results, which demonstrates its value in practical application.(3) A novel water-soluble fluorescent probe 3,4 – boronate –N-(2- pyridylmethyl)-1,8- naphthalimide ethyl iodide quaternary ammonium salt, for detection of hydrogen peroxide was designed and synthesized. The probe itself had weak fluorescence, it would react with hydrogen peroxide that was produced by catalyzing acetylcholine to produce the fluorescent product, and resulted in an increase in fluorescence, so the concentration of ACh could be detected. Researches showed the method for detection of ACh had the advantage of excellent selectivity, high sensitivity, simple operation and less time consuming, meanwhile, the strategy could be used to monitor pesticide residue in food and had high application value.
